Nitrogenase is the only enzyme that catalyzes the reduction of nitrogen gas into ammonia. Nitrogenase is tightly inhibited by the environmental gas carbon monoxide (CO). Many nitrogen fixing bacteria protect nitrogenase from CO inhibition using the protective protein CowN. This work demonstrates that a conserved glutamic acid residue near the C-terminus of Gluconacetobacter diazotrophicus CowN is necessary for its function. Mutation of the glutamic acid residue abolishes both CowN’s protection against CO inhibition and the ability of CowN to bind to nitrogenase. In contrast, a conserved C-terminal cysteine residue is not important for CO protection by CowN. Overall, this work …

Abstract. This research used deep learning for image analysis by isolating and characterizing distinct DNA replication patterns in human cells. By leveraging high-resolution microscopy images of multiple cells stained with 5-Ethynyl-2′-deoxyuridine (EdU), a replication marker, this analysis utilized Convolutional Neural Networks (CNNs) to perform image segmentation and to provide robust and reliable classification results. First multiple cells in a field of focus were identified using a pretrained CNN called Cellpose. After identifying the location of each cell in the image a python script was created to crop out each cell into individual .tif files. After careful annotation, a CNN was …

Background; Physical activity after surgical resection for colon cancer is associated with significantly longer disease-free survival. Inflammation is hypothesized to mediate the association between physical activity and disease-free survival in colon cancer. Methods; In this exploratory analysis of a randomized dose-response trial, 39 colon cancer survivors who completed standard therapy were stratified by cancer stage and randomized in a 1;1;1 ratio to one of three treatment groups for 24 weeks of usual-care control, 150 min/wk of moderate-intensity aerobic exercise (low-dose), or 300 min/wk of moderate-intensity aerobic exercise (high-dose). The field of shark paleoecology often yields indecisive conclusions based on the limited fossilization of their anatomical structures, with the exception of their teeth. The majority of the Atlantic coast has been studied regarding the presence of certain prehistoric shark species from the Miocene, Pliocene, and Pleistocene epochs. However, information pertaining to the Georgia coast and understanding its potential community structure is relatively understudied. Mitochondrial-targeting therapy aims to restore or enhance mitochondrial function to treat or alleviate these conditions. To determine the free-radical scavenging activity and total phenolic content of the ethanol extract of Poinsettia leaves (Euphorbia pulcherrima Willd.) from Lombok Island, we used the 1,1-diphenyl-2-picrylhydrazyl method and expressed the total phenolic content as gallic acid equivalent (GAE) per gram of ethanol extract. The results show the strong antioxidant activity (79.77 ppm) of the ethanol extract of red Poinsettia leaves and the moderate antioxidant activity of green Poinsettia leaves (118.350 ppm). The ethanol extracts of red and green Poinsettia leaves possessed an average of 63.276 ± 0.228 and 22.715 ± 0.090 mg GAE/g extract total phenolic contents, respectively. …

In this study, a screen-printed electrode (SPE) for nitrite (NO2) sensing was fabricated through an inkjet printing technique using a commercial printer machine and preparing an ink composite solution containing graphene oxide (GO) and poly(3,4 ethylenedioxythiophene) poly(styrene sulfonate) (PEDOT:PSS) as conductive materials. The optimum ink materials for SPE fabrication comprised GO and PEDOT:PSS with binders that resemble the viscosity of commercial ink. This study was guided by the central research question: What are the lived experiences and perceptions of former African American teachers and students that endured segregation, desegregation, and integration in Arkansas' Delta? The phenomenological study used eight former African American teachers and student participants. Of the participants, there were four former teachers and four former students. In this study, the participants' voices are captured as they share their experiences and perception of each phase of education that influenced their lives and left an everlasting imprint. Oyster reefs provide a variety of important ecosystem services. However, the mortality rate of eastern oyster, Crassostrea virginica, the dominant species that produces oyster reefs in the northern Gulf of Mexico, is increasing at an alarming rate due to a variety of abiotic and biological factors. I examined how biophysical factors, including the less-studied fatty acid profiles of the suspended particulate matter on which oysters feed, influenced morphometric condition of C. virginica.

A key question in understanding the implications of climate change is how to integrate ecological risk assessments that focus on contaminants with the environmental alterations from climate projections. Universal phylogenetic markers such as the nuclear ribosomal internal transcribed sequences (ITS), specifically ITS1 and ITS2, are routinely used to estimate fungal diversity in environmental samples. However, numerous studies report differences in the performance and efficacy of ITS1 and ITS2 in documenting fungal diversity. Examining the activity patterns of wildlife is an important aspect of understanding the ecology of a species and may be especially important for species of conservation concern. We used remotely triggered cameras to describe the daily and seasonal activity patterns and examine ecological factors that influence the activity of the Amargosa Vole (Microtus californicus scirpensis), a California endemic listed federally and by the state as Endangered, and is a marsh habitatspecialist in the Mojave Desert. Crabgrass (Digitaria spp.) is an annual summer grass that can provide high-quality forage, but optimal management strategies are unclear. Our objective was to compare the yield and quality of crabgrass (Mojo and Quick-N-Big) under different nitrogen rates and harvest management. The experimental design was a randomized complete block with five treatments and three replications for each crabgrass variety, totaling fifteen experimental units for both Mojo and Quick-N-Big, in adjacent sites. The objective of this study was to evaluate the chemical composition and microbial diversity of the silage of forage peanut harvested at 40 and 80 days of regrowth and treated with microbial inoculants. The inoculants evaluated were commercial inoculant (CI) Sil-All 4×4 (Lallemand®, Brazil); the AV14.17, Lactiplantibacillus pentosus strain (ISO) isolated from alfalfa silage; and control (CTRL), without inoculant. In the context of reducing greenhouse gas (GHG) emissions to combat climate change, rewetting of peatlands is of particular importance. High groundwater levels covering organic matter in the soil prevent its oxidation to CO2. The amount of water needed to allow the rewetting of peatlands can be determined in lysimeter studies.

In an attempt to estimate forage production and carrying capacity of wild ungulates in Tandoureh National Park of Iran, the Babanestan grassland was selected, as the typical habitat of about 200 Urial wild sheep. In order to estimate the carrying capacity of this habitat for foraging of Urial wild sheep, this habitat with an area of 132 hectares was classified as low flat (LF), high flat (HF), north aspect (NA), and south aspect (SA). Interspecific hybrids of genus Pennisetum (P. glaucum x P. purpureum) is the one of the most popular manmade hybrid. It combines the unique features of both P. glaucum (Pearl millet; Bajra) and P. purpureum (Napier; Elephant grass) species, which makes it more resilient to harsh environments with superior fodder quality. In grass-legume pastures, 2,4-D-amine + florpyrauxifen-benzyl controls broadleaf weeds but red clover (Trifolium pratense L.) is eliminated. Grazing within a week prior to herbicide application is likely to reduce leaf surface area and may reduce red clover injury and maintain productivity. Experiments were conducted in 2020 and in 2021 to determine if red clover could be productive when 560 g ae ha-1 2,4-D-amine + 6.3 g ae ha-1 of florpyrauxifen-benzyl with 1% v/v methylated seed oil applied in the spring was grazed before or after herbicide application. Paspalum dilatatum Poir. (dallisgrass) is an excellent C4 forage for summer production, highly productive with a wide distribution within warm-temperate regions. Tropical grasses have characteristics that allow the occurrence of undesirable fermentations resulting in low quality silage. Inoculants can improve silage quality.
