The oncogenic corepressors C-terminal Binding Protein (CtBP) 1 and 2 harbor regulatory d-isomer specific 2-hydroxyacid dehydrogenase (d2-HDH) domains. 4-Methylthio 2-oxobutyric acid (MTOB) exhibits substrate inhibition and can interfere with CtBP oncogenic activity in cell culture and mice. Crystal structures of human CtBP1 and CtBP2 in complex with MTOB and NAD(+) revealed two key features: a conserved tryptophan that likely contributes to substrate specificity and a hydrophilic cavity that links MTOB with an NAD(+) phosphate. Neither feature is present in other d2-HDH enzymes. These structures thus offer key opportunities for the development of highly selective anti-neoplastic CtBP inhibitors. Elsevier B.V. All …

Under the selective pressure of therapy, HIV-1 protease mutants resistant to inhibitors evolve to confer drug resistance. Such mutations can impact both the dynamics and structures of the bound and unbound forms of the enzyme. Flap+ is a multidrug-resistant variant of HIV-1 protease with a combination of primary and secondary resistance mutations (L10I, G48V, I54V, V82A) and a strikingly altered thermodynamic profile for darunavir (DRV) binding relative to the wild-type protease. We elucidated the impact of these mutations on protein dynamics in the DRV-bound state using molecular dynamics simulations and NMR relaxation experiments. The usage of 128 bit addresses with hexadecimal representation in IPv6 poses significant potential privacy issues. This paper discusses the means of allocating IPv6 addresses, along with the implications each method may have upon privacy in different usage scenarios. The division of address space amongst the global registries in a hierarchal fashion can provide geographical information about the location of an address, and its originating device. ZnO nanorods were grown on a variety of substrates such as Si, SiO2/Si and sapphire in a large-area pulsed laser deposition chamber designed for sensor device fabrication. Processing conditions were optimized to grow ZnO nanorods with or without seed layers. Au, Cr and BaSrTiO3 (BST) seed layers were investigated to compare their effects on the diameter and orientation of ZnO nanorods. ZnO nanorods were observed to align better when grown on sapphire, Cr or BST seed layers as compared to Au or Si layers. Longitudinal stream profiles provide valuable information concerning geomorphic features and energy states in a stream. Traditionally, stream profiles have been generated by field surveying or topographic map analysis. The continued growth of digital data and Geographic Information Systems (GISs) provide another method by which to achieve stream profile generation. This work examines the effectiveness of digital data, digital elevation models (DEM), and GIS to construct stream profiles. To determine the most effective and accurate data for profile generation, profiles were created using 1-meter (1-m) and 3-meter (3-m) DEMs developed from LiDAR data. The pKa of a compound is one of its most important properties as it defines the specific molecular forms that will prevail under different pH conditions. Accordingly, accurate means for computational estimation of this property are of particular interest. The complex drainage systems within karst settings can result in atypical longitudinal profiles. Features, such as cave entrances, can be expressed as anomalous ‘bumps’ in the longitudinal profile of a stream if downcutting has continued upstream of the area in which the water is pirated to the subsurface. Horn Hollow, a fluviokarst valley located in Carter Caves State Park Resort in northeastern Kentucky, was examined for these types of features. ZIP transporters, named after the zinc regulated (Zrt) and iron regulated (Irt) transport proteins, are essential for zinc and iron translocation across cellular membranes. These proteins function to increase the cytosolic concentration of transition metals. While both zinc and iron are essential micronutrients which are required for the structure and/or function of hundreds of cellular proteins, the molecular mechanism of ZIP transporters is not well understood. Complicating mechanistic studies is the observation that the concentration of free zinc and iron is nano to picomolar.

This paper presents an implantable polymer/metal alloy thin film structure for localized post-operative treatment of breast cancer. A combination of experiments and models is used to study the temperature changes due to Joule heating by patterned metallic thin films embedded in poly-dimethylsiloxane. The heat conduction within the device and the surrounding normal/cancerous breast tissue is modeled with three-dimensional finite element method (FEM). The FEM simulations are used to explore the potential effects of device geometry and Joule heating on the temperature distribution and lesion (thermal dose). Maximizing resource extraction from mature oilfields requires enhanced and secondary recovery techniques. The success of these methods relies on knowledge and understanding of the reservoir geology and hydraulics. At the Loudon Oilfield (Illinois, USA), enhanced oil recovery is being used to extend the production life of the reservoir. The suitability and placement of additional wells for oil recovery processes required three-dimensional (3D) facies and porosity modeling of the oilfield. In order to study the economic impact of an earthquake prediction system, we use probabilistic methods to model the expected cost per life saved from a prediction system. We improve upon previous work by directly modeling the expected cost per life saved rather than using the ratio of the expected cost to the expected number of lives saved, which we show is always an underestimate.  The model is applied numerically to the San Francisco Bay area and the expected cost per life saved from an earthquake prediction system over a 50 year period is found to be $3.3 million.  While …

Natural gas is stored underground in geologic structures throughout the United States. However, complexities associated with these geologic structures may provide vertical pathways for gas migration, and thus gas loss. Possible upward migration (loss) of natural gas in an underground gas storage field in stimulated this investigation that aims to identify potential migration pathways. Spatial analysis of volume of shale (Vsh) and formation porosity (n) values were conducted in conjunction with high-resolution shallow seismic surveys to identify potential vertical pathways. In this paper we review important aspects of semiparametric theory and empirical processes that arise in causal inference problems. We begin with a brief introduction to the general problem of causal inference, and go on to discuss estimation and inference for causal effects under semiparametric models, which allow parts of the data-generating process to be unrestricted if they are not of particular interest (i.e., nuisance functions).
