Two application areas, namely language learning and speech recognition adaptation, are largely driving this research interest and are the focal points of this work.

Over the course of the last century, we have observed changes in forest composition and structure related to changes in disturbance regimes. Reduced frequency and severity of disturbance events has allowed for the ingrowth of shade-tolerant, mesophytic species into forest understories; the result is shifting species compositions and changes in forest structure. In some cases, the loss of masting species, such as oak (Quercus spp.) and hickory (Carya spp.), may also accompany a shift in forest function and further accommodate other species shifts among mammal, insect, and bird populations. Covariance functions play a central role in spatial statistics. Parametric covariance functions have been used in most of the existing works on the analysis of spatial data. The primary reason for this is that the classes of parametric covariance functions guarantee that the fitted covariance function is positive definite. In this dissertation, I undertake two non-parametric approaches to modelling the covariance functions.

A new instrument incorporating ion trap based tandem mass spectrometry and cold ion UV and UV-IR double resonance spectroscopy has been developed at Purdue University for the study of gas phase, biologically relevant ions. The instrument incorporates multiple quadrupole linear ion trap mass analyzers to prepare and isolate the precursor ions as well as to analyze the resultant photofragments. A 22-pole ion trap cooled to 5 K via a closed cycle helium cryostat is used to cool the ions for spectroscopic interrogation. Results show the vibrational and rotational temperature of a wide assortment of ions to be 12±2 K.

Enantioselective separations of chiral molecules are important in various chemical fields, such as pharmaceuticals and agrochemicals industries. Polysaccharide-based sorbents have been widely used in chiral liquid chromatography. The recognition mechanisms which determine their enantioselectivities are not completely understood.

In this dissertation, the chiral recognition mechanisms of a widely used commercial sorbent, amylose tris[(S)-alpha-methylbenzylcarbamate], for benzoin (B) enantiomers were first studied. The HPLC data for benzoin with pure n-hexane as the mobile phase have been obtained. The behavior of sorbent-solute-hexane systems can be interpreted by considering only sorbent solute two-component interactions. The work in this thesis details the design, synthesis, and biological evaluation of molecular inhibitors for the inhibition of biologically relevant enzymes. The first three chapters of this thesis concern the polyphenol resveratrol and its inhibition of the quinone reductase 2 (QR2) enzyme. The work on this subject resulted in the complete design, synthesis, biological and structural evaluation of a second generation library of resveratrol analogues. This research uses molecular dynamics, kinetic Monte Carlo and binary collision approximation methods to model fusion relevant material bombarded by energetic particles to investigate retention, deposition, sputtering, erosion, blistering effects, diffusion, and so on.

The deuterium bombardment of monocrystalline tungsten was modeled by LAMMPS code using Tersoff type interatomic potential. Various aspects of the interaction of low-temperature plasma (LTP) ambient ionization with organic molecules and surfaces are discussed. As an application, LTP-MS has been used for the analysis of pesticides directly from fruits and vegetables revealing low parts per billion limits of detection (LODs). Due to low power and gas requirements, a battery-operated and handheld LTP probe was developed for the point-and-shoot MS analyses. Handheld LTP was compared with large-scale LTP on a benchtop and miniature MS revealing similar LODs for both. The detection of GeV/TeV emission from X-ray binaries (XRBs) has established a new class of high-energy (HE, >0.1 GeV) and very-high-energy (VHE, >100 GeV) gamma-ray emitters. XRBs are formed by a compact object, either a neutron star or a black hole, and by an optical companion star. Some XRBs are known to possess collimated relativistic jets, and are called microquasars. VERITAS has conducted observations of the high-mass X-ray binary (HMXB) 1A 0535+262 and of the microquasar Cygnus X-3. During this gas-phase chemistry, particle transfer (i.e., proton and electron) or synthesis can occur at rapid reaction rates. Particle transfer represents a mature area of ion/ion chemistry, while selective covalent modification represents a fairly new area of gas-phase chemistry. Nitrogen (N) is the most limiting essential nutrient for crop growth. Numerous studies have been performed to improve N fertilizer recommendations. Accurate prediction of soil N supply has been found to be one of the most important factors that determine optimum fertilizer N rates. Seven soils collected from various locations across Indiana at four different depths were tested for N mineralization potential. Results showed that laboratory mineralizable N in the top layer (0-15 cm) of the seven soils ranged from 50 to 68 mg N kg-1 soil. Metabolic reprogramming has been increasingly recognized as a core hallmark of cancer. My dissertation work identified novel diagnostic markers and therapeutic targets for human cancers through the study of cholesterol in cancer cells.

Enabled by label-free Raman spectromicroscopy, we performed the first quantitative analysis of lipogenesis at single cell level in human patient cancerous tissues. This dissertation argues that a new paradigm of selfhood emerged in nineteenth-century British literature, one that recognized the individual will and environmental influence not as antithetical but as dialectical forces in the formation of the self. The concept of an externally negotiated subject challenges both the inward and socially determined conceptions of self that have dominated the relevant criticism.
