This research contains two topics: (1) PBNPA: a permutation-based non-parametric analysis of CRISPR screen data; (2) RCRnorm: an integrated system of random-coefficient hierarchical regression models for normalizing NanoString nCounter data from FFPE samples.
Clustered regularly-interspaced short palindromic repeats (CRISPR) screens are usually implemented in cultured cells to identify genes with critical functions. Although several methods have been developed or adapted to analyze CRISPR screening data, no single spe- cific algorithm has gained popularity. Thus, rigorous procedures are needed to overcome the shortcomings of existing algorithms. The Bernoulli distribution is a basic, well-studied distribution in probability. In this thesis, we will consider repeated Bernoulli trials in order to study runs of identical outcomes. More formally, for t ∈ N, we let Xt ∼ Bernoulli(p), where p is the probability of success, q = 1 − p is the probability of failure, and all Xt are independent. The focus of this thesis was to investigate which baseball metrics are most conducive to run creation and prevention. Stepwise regression and Liu estimation were used to formulate two models for the dependent variables and also used for cross validation. Finally, the predicted values were fed into the Pythagorean Expectation formula to predict a team’s most important goal: winning.

Multiple regression models play an important role in analyzing and making predictions about data. Prediction accuracy becomes lower when two or more explanatory variables in the model are highly correlated. One solution is to use ridge regression. The purpose of this thesis is to study the performance of available ridge regression estimators for Poisson regression models in the presence of moderately to highly correlated variables. As performance criteria, we use mean square error (MSE), mean absolute percentage error (MAPE), and percentage of times the maximum likelihood (ML) estimator produces a higher MSE than the ridge regression estimator. Despite efforts to increase participation of racial and ethnic minorities (excluding Asians) in science, technology, engineering and mathematics (STEM) in the United States, this group remains underrepresented in these fields. Many efforts to increase minority participation focus on support structures to help this group “get through” the pipeline. However, less attention has been paid to increasing their intrinsic motivation to pursue careers in STEM. Our work is focused on increasing this intrinsic motivation, looking at role models as external influences. Analysing linked, routinely collected data may be useful to identify characteristics of patients with suspected lung cancer who could benefit from early assessment for palliative care. The aim of this study was to compare characteristics of newly diagnosed lung cancer patients dying within 30 days of diagnosis (short term survivors) with those surviving more than 30 days. To identify indicators for early palliative care assessment we distinguished between characteristics available at diagnosis (age, gender, smoking status, marital status, comorbid disease, admission type, tumour stage and histology) from those available post diagnosis. Statistical association is an important concept in statistics. An exploratory study examined how students reason about statistical association utilizing graphical representations constructed with CODAP, a dynamic statistical graphing software. Task-based interviews were conducted with three 6th grade students prior to formal instruction. Students’ conceptions of a statistical relationship, proportional reasoning skill level, ability to interpret bivariate categorical graphs (particularly segmented bar graphs and two-way binned plots), and ability to identify association of two categorical variables were all investigated through interview tasks and responses to inquiry. Relationships between the Madden–Julian oscillation (MJO) and the extratropical circulation have been leveraged extensively to improve subseasonal prediction. However, in certain situations, tropical cyclones (TCs), which often coincide with enhanced MJO activity, can constructively or destructively interfere with MJO organization and common pathways through which the extratropics responds to the MJO. To examine this potential interference, the first portion of this study leverages a statistical experiment to relate West Pacific TC presence in different parts of the basin during a given MJO phase to subsequent remote extratropical circulation outcomes.
