Divergence, Recombination And Retention Of Functionality During Protein Evolution, Yanong O. Xu, Randall W. Hall, Richard A. Goldstein, David D. Pollock

Collected Faculty and Staff Scholarship

We have only a vague idea of precisely how protein sequences evolve in the context of protein structure and function. This is primarily because structural and functional contexts are not easily predictable from the primary sequence, and evaluating patterns of evolution at individual residue positions is also difficult. As a result of increasing biodiversity in genomics studies, progress is being made in detecting context-dependent variation in substitution processes, but it remains unclear exactly what context-dependent patterns we should be looking for. To address this, we have been simulating protein evolution in the context of structure and function using lattice models …

Ronald Cerny Publications

Histone Lys methylation plays an important role in determining chromatin states and is mostly catalyzed by SET domain– containing proteins. The outcome, transcriptional repression or activation, depends on the methylated histone residue, the degree of methylation, and the chromatin context. Dimethylation or trimethylation of histone H3 Lys 4 (H3K4me2 or H3K4me3) has been correlated with transcriptionally competent/active genes. However, H3K4 methylation has also been implicated in gene silencing. This dualistic nature of the H3K4 methyl mark has thus far remained unresolved. In the green alga Chlamydomonas reinhardtii, Mut11p, related to a subunit of trithorax-like methyltransferase complexes, is required for transcriptional …

Synthesis Of Lanthanum-Strontium Magnanites By A Hydroxide-Precursor Co-Precipitation Method In Solution And In Reverse Micellar Microemulsion, Vuk Uskoković, Darko Makovec, Miha Drofenik

Pharmacy Faculty Articles and Research

Nanostructured lanthanum-strontium manganites have been synthesized using two different co-precipitation approaches, one in bulk solution, and the other in reverse micelles of CTAB/1-hexanol/1-butanol/water microemulsion. In both cases, precursor cations were precipitated by alkali precipitating agents. The properties of the material synthesized by using these two methods were compared in order to reveal potential advantages of microemulsion-assisted approach. The influence of the annealing conditions on the properties of synthesized manganites was investigated by using X-ray diffraction, transmission electron microscopy, differential thermal analysis, thermogravimetric analysis and magnetic measurements.

A Mechanism For The Formation Of Nanostructured Nizn Ferrites Via A Microemulsion-Assisted Precipitation Method, Vuk Uskoković, Miha Drofenik

Pharmacy Faculty Articles and Research

Nanostructured NiZn ferrites were synthesized using two different techniques: first, a precipitation procedure in the reverse micelles of a CTAB/1-hexanol/H2O microemulsion, and second, precipitation in a bulk aqueous solution. XRD measurements, magnetic measurements, TEM imaging, analytical measurements and thermal analyses were used in an attempt to reveal the chemical pathway that leads to the formation of NiZn ferrite in the microemulsion and in the bulk aqueous solution. It was found that reverse micelles do not act as inert nano-sized reactors that influence only morphological properties of the synthesized powders, but have a decisive influence on the identity of …

Effect Of Melatonin And Dopamine In Site Specific Phosphorylation Of Phosducin In Intact Retina, Arinzechukwu Okere Nkemdirim

Theses and Dissertations

Phosducin (Pdc) is a 28 kDa binding partner for the G protein beta gamma subunit dimer (G-beta-gamma) found abundantly in the photoreceptor cells of the retina and pineal gland. In the retina, light-dependent changes in cAMP and Ca2+ control the phosphorylation of Pdc at serine 73 and 54, respectively, which in turn controls the binding of Pdc to G protein beta gamma subunit dimer . G protein beta gamma subunit dimer binding has been proposed to facilitate light-driven transport of G protein beta gamma subunit dimer from the site of phototransduction in the outer segment of the photoreceptor cell to …

Nafion Emulsion Method For Fuel Cell Membrane-Electrode-Assembly Preparation, Chao-Yang Xia, Lin Zhuang

Journal of Electrochemistry

A Nafion emulsion method for fuel cell membrane-electrode-assembly (MEA) preparation was reported for the first time in this paper. 5 %(by mass) Nafion alcohol solution and non-polar solvent (e.g., cyclohexane) were ultrasonicallymixed to produce Nafion emulsion. The catalyst ink prepared using the thus-obtained Nafion emulsion can be directly cast onto Nafion membrane to make MEA. This method is simple in operation, and effectively preventsthe Nafion membrane swelling, hence is suitable for large-scale application. The MEA thus-prepared showed comparable catalyst efficiencywith respect to the popular transfer-method, and the preliminary performance in room-temperature H_(2)/O_(2 )fuel cell test was satisfying.

A Preliminary Study Of Influence Of The Chain Length Of Alkanethiols On The Electrochemical Behavior Of Redox Self-Assembled Multilayer Thin Films, Yun-Feng Wang, Zhan-Jun Zhang

Journal of Electrochemistry

Alkanethiols and ferrocene derivates were used to construct redox self-assembled double-or triplex-layer thin films, which acted as a model of redox self-assembled multilayer thin films (SAMTFs) to study the in-fluence of alkanethiols chain length on the electrochemical behavior of the SAMTFs. The rate of electron transferreaction decreased exponentially with the distance between the Fc groups and the underlying electrodes. The in-fluence of chain length on the electrochemical behaviors of the SAMTFs was inconsistent under different situa-tions. On the experimental situation of this work, when electroactive groups were closer to the electrodes,SAMTFs by longer chains of alkanethiols displayed stronger electrochemical responses, …

Surface-Enhanced Raman Spectra Of Cytosine Adsorbed On A Roughened Gold Electrode, Yuan-Fei Wu, Jian-Feng Li, De-Yin Wu, Bin Ren, Zhong-Qun Tian

Journal of Electrochemistry

Cytosine adsorbed on a roughened gold electrode has been investigated by using electrochemical cyclic voltammetry and surface-enhanced Raman spectroscopy (SERS). The results indicate that cytosine was adsorbed on the Au electrode in a perpendicular orientation via the N3 position. As the maximum enhancement of ring breathing mode appears at negative potentials compared with other modes, both the electromagnetic enhancement and the charge transfer mechanism were considered to contribute to the SERS intensity. The binding interaction between cytosine and the gold electrode was weakened when the potential was negatively moved. The result also indicates that the SERS technique can be used …

A Conjugate Dissolution / Deposition Behavior In Thin Layer Electrodeposition, Sun Bin Aug 2005

A Conjugate Dissolution / Deposition Behavior In Thin Layer Electrodeposition, Sun Bin

Journal of Electrochemistry

During the electrodeposition of lead, the dendrite deposit segregates from the cathode due to the disturbance of hydrogen gas. The segregated deposit could move toward the anode spontaneously rather than keep stationary. The studies show that this novel phenomenon was caused by the conjugate electrodissolution and electrodeposition reactions taking place at two ends of the isolated segment of the deposit. The mechanism inducing this phenomenon is suggested.
