Waste streams of hydrogen and helium mixtures are produced at the Kennedy Space Center during purging of the hydrogen systems and supply lines. This process is done prior to and after hydrogen servicing. The purged waste gases are lost to the atmosphere, resulting in an annual loss of 2 million and 0.1 million standard cubic meters of helium and hydrogen, respectively. Recovery of these gases will have an economic benefit. Metals, alloys, and intermetallics are known to react with hydrogen in favorable conditions; therefore, they have the possibility of serving as separating and recovery agents. Conventional methods of body fluid identification use labor-intensive, technologically diverse techniques that are performed in a series, not parallel, manner and are costly in terms of time and sample. Furthermore, for some frequently encountered body fluids, such as saliva or vaginal secretions, no confirmatory technique exists. Terminally differentiated cells, such as blood lymphocytes or epithelial cells lining the oral cavity, have a unique pattern of gene expression, which is evinced by the presence and relative abundance of specific mRNA species. The thermal degradation of polycarbonate/triphenylphosphate (PC/TPP) and PC/resocinolbis(diphenylphosphate) (PC/RDP) in air has been studied using TGA/FTIR and GC/MS. In PC/phosphate blends, the phosphate stabilizes the carbonate group of polycarbonate from alcoholysis between the alcohol products of polycarbonate degradation and the carbonate linkage. Thus, the evolution of bisphenol A, which is mainly produced via hydrolysis/alcoholysis of the carbonate linkage, is significantly reduced, while, the evolution of various alkylphenols and diarylcarbonates increases. The bonds that are broken first in the thermal degradation of both the carbonate and isopropylidene linkages of polycarbonate are the weakest bonds in each, when a phosphate is present. …

We report an extensive study on total and partial-ion-yield spectroscopy around both the S 2p and F 1s thresholds in SF6. All positive and negative single-ion channels have been measured. Below the F 1s threshold we detect a large variation in relative intensity of the resonant structures according to the specific channel monitored, indicating selective fragmentation. Above threshold, at variance with previous cases described by us, we detect high-intensity structures related to shape resonances not only in the cation channels but also for the anions. The structural and electronic properties of Gd2(Ti1−yZry)2O7 (y=0–1) pyrochlores following a 2.0-MeV Au2+ ion-beam irradiation (~5.0 X 1014 Au2+/cm2) have been investigated by Ti 2p and O 1s near-edge x-ray absorption fine structure (NEXAFS). The research in this dissertation focuses on the design of a novel redox-responsive host system that has the ability to encapsulate guest molecules. The host at hand is a tri(ethylene oxide), TEO, pyrrole-terminated PPI dendrimer that possesses water-solubility as a result of the presence of the TEO chains on the pyrrole groups and redox-responsive activity from the pyrrole groups after their oligomerization. The pyrrole moieties at the periphery of the dendrimer exhibit the ability to be oligomerized by oxidation chemistry, and this is shown to aid in the retention and egress of guest molecules located within the dendrimer interior. The Reno Aerosol Optics Study (RAOS) was designed and conducted to compare the performance of many existing and new instruments for the in situ measurement of aerosol optical properties with a focus on the determination of aerosol light absorption. For this study, simple test aerosols of black and white particles were generated and combined in external mixtures under low relative humidity conditions and delivered to each measurement system. The aerosol mixing and delivery system was constantly monitored using particle counters and nephelometers to ensure that the same aerosol number concentration and amount reached the different instruments. The release of the 1000th complete microbial genome will occur in the next two to three years. In anticipation of this milestone, the Fellowship for Interpretation of Genomes (FIG) launched the Project to Annotate 1000 Genomes. The project is built around the principle that the key to improved accuracy in high through put annotation technology is to have experts annotate single subsystems over the complete collection of genomes, rather than having an annotation expert attempt to annotate all of the genes in a single genome. In this study, direct measurements of the air/ sea flux of acetone were made over the North Pacific Ocean. The results demonstrate that the net flux of acetone is into, rather than out of the oceans. The extrapolated global ocean uptake of 48 Tg yr(-1) requires a major revision of the atmospheric acetone budget. This result is consistent with a recent reevaluation of acetone photodissociation quantum yields.

The synthesis of amide-linked disaccharide mimetics has been explored starting with carbohydrate-based amines and a protected quinic acid lactone. Benzyl-2-amino-4,6-Obenzylidene-2-deoxy-α/β-D-glucopyranose (12) and D-glucamine (14) were successfully coupled to give the corresponding quinamides (13 and 15), while the quinoylation of Oacetylated L-fucopyranosyl methylamine (7) failed. The latter was prepared from per-O-acetylL-fucopyranose via the improved multigram scale synthesis of the corresponding per-O-acetylL-fucopyranosyl cyanide (3). Compound 3 was subsequently hydrogenated to yield a mixture of compound 7 and the per-O-acetylated bis-(fucopyranosylmethyl) amine (5). The occurrences of two recurrent motifs in ribosomal RNA sequences, the Kink-turn and the C-loop, are examined in crystal structures and systematically compared with sequence alignments of rRNAs from the three kingdoms of life in order to identify the range of the structural and sequence variations. Isostericity Matrices are used to analyze structurally the sequence variations of the characteristic non-Watson-Crick base pairs for each motif. We show that Isostericity Matrices for non-Watson-Crick base pairs provide important tools for deriving the sequence signatures of recurrent motifs, for scoring and refining sequence alignments, and for determining whether motifs are conserved throughout evolution. …

The synthesis of the new aryl bis(β-diketone) ligand m-phenylenebis(acetylacetone) (m-pbaH2) allowed for complexation with copper(II) to form a 4-membered ring Cu4(m-pba)4. An alternate route, via a phospholene adduct, was implemented after difficulties in preparing the ligand via an azo sulfide conversion from m-phenylenediamine. The connection of two m-pba ligands coordinated to a Cu(II) metal ion causes a distortion along the axial position of the metal center. The pore has a diameter of (Cu-----Cu = 14.317 Å) when viewing opposite metal centers. An open coordination site along Cu(II) axial position may facilitate host-guest binding interactions. In oncologic applications of photodynamic therapy (PDT), the discriminating localization of porphyrin-type compounds in solid tumors is exploited for the selective ablation of neoplastic tissue with minimal destruction and irritation to normal tissue. PDT is a locoregional, binary cancer therapy in which a photosensitizer—light-activated drug—absorbs light of an appropriate wavelength and excites to the singlet state. This photosensitizer in the excited singlet state can undergo an internal transition to the excited triplet state, a relatively long-lived and high-energy species that transfers its excess energy to molecular oxygen. The existence of shape selectivity in non-covalent molecularly imprinted polymers has been proven using molecular probes. A series of amines varying with different structural motifs and secondary amines with different sized side chains were imprinted, and binding evaluated by HPLC for each amine on polymers imprinted with similar amines. Trends in the binding relationships revealed two major contributions of cavity structure on selectivity afforded by molecularly imprinted polymers (MIPs). The influence of shear on viscoelastic solutions of poly(ethylene oxide) (PEO) and clay (Cloisite, CNA) was investigated by rheology and small angle neutron scattering (SANS) under shear. These measurements determined the shear-induced orientation of the clay and the polymer as well as their relaxation behavior after cessation of shear. Comparison of PEO-CNA solutions (~100 nm diameter platelets) to previously studied PEO-Laponite gels (~30 nm diameter platelets) found that the orientation of CNA platelets occurs at much lower shear rates. Additionally, the relaxation times were much longer for CNA platelets than for Laponite platelets. From these solutions and gels, multilayered nanocomposite …

The goal of the research presented in this dissertation is to develop effective analytical methods for the separation of both achiral and chiral analytes using polymeric surfactants. Three analytical techniques are employed to accomplish this objective: open-tubular capillary electrochromatography (OT-CEC), open-tubular capillary electrochromatography coupled to mass spectrometry (OT-CEC/MS), and micellar affinity gradient focusing (MAGF). The scope of this dissertation is to explore the enantiomeric recognition properties of amino acid based polymeric surfactants using a combination of analytical techniques including chromatographic and spectroscopic methods. Chapter 1 includes an introduction to chirality and chiral separations using capillary electrophoresis. Using cyclodextrin modified micellar electrokinetic chromatography (CD-MEKC), the enantioseparation of three binaphthyl derivatives using native alpha- and beta-cyclodextrins in combination with various diastereomers of chiral polymeric surfactants (PS) was examined. Polychlorinated dibenzo-p-dioxins and dibenzofurans (PCDD/F) are toxic compounds produced in combustion systems such as incinerators, and their emissions are an important environmental issue. Chlorinated phenols and benzenes form PCDD/F through heterogeneous reactions following adsorption onto fly-ash surfaces. The goal of the research presented in this dissertation is to synthesize, characterize, and apply a novel chiral amino acid-based surfactant and achiral sulfate-based surfactants for the effective separations of chiral and achiral compounds in micellar electrokinetic chromatography (MEKC). The first part of this research involves novel synthesis, characterization, and application of polysodium oleyl-L-leucyl-valinate (poly-L-SOLV) for the separation of chiral compounds in MEKC. Surface tensiometry, proton nuclear magnetic resonance, infrared spectroscopy, densitometry and analytical ultracentrifugation were used as the characterization techniques. As technology begins to utilize nanocrystals for many chemical, biological, medical, electrical, and optoelectrical applications, there is a growing need for an understanding of their fundamental properties. The study of melting and solidification of nanocrystals is of interest to fundamental understanding of the effect of reduced size and crystal shape on the solid-liquid phase transition. Melting and solidification of as-deposited and recrystallized indium and bismuth nanocrystals were studied using reflection high-energy electron diffraction (RHEED). Novel symmetric and asymmetric water-soluble tetrabenzoporphyrin are synthesized in relatively good yields. These compounds are synthesized by the condensation of butanopyrrole with several aldehydes that contain masked water solubilizing groups followed by metal insertion, oxidation and demetallation and deprotection of the masked solubilizing groups. The biological evaluation (in vitro and in vivo) studies are described in addition to the photophysical evaluations of the corresponding compounds. A novel versatile route to the syntheses of novel free-bases and metallo-benzoylbiliverdin is discussed. Research, inspired by nature, provides ample opportunity for discovery, ingenuity, and creative endeavor. For these reasons, the work and accomplishments presented herein focus on the total syntheses and related investigations of a variety of natural product derivatives, specifically (+)-nootkatone and jasmone. While the former consists of several independent projects: (a) an enantioselective synthetic route to (+)-nootkatone and derivatives, (b) examination of the valencane ring system, (c) determination of the absolute configuration of (+)-nootkatone, and (d) rearrangements of various sesquiterpenoid derivatives; the latter focuses on the construction ofƒndelta, epsilon-unsaturated carbonyl compounds, hetero Pauson-Khand substrates, via [3,3]-sigmatropic rearrangements.

Increasing the information content obtainable from a single assay and system miniaturization has continued to be important research areas in analytical chemistry. The research presented in this dissertation involves the development of a two-color, time-resolved fluorescence microscope for the acquisition of both steady-state and time-resolved data during capillary and microchip electrophoresis. The utility of this hybrid fluorescence detector has been demonstrated by applying it to DNA sequencing applications. Coupling color discrimination with time-resolved fluorescence offers increased multiplexing capabilities because the lifetime data adds another layer of information. Anions play important roles in biological systems. The design of anion sensors has been of great interest. The selective detection of a specific anion is particularly challenging. In the first chapter, I describe the synthesis of a TBS-protected fluorescein dye (1.1) and its use for the selective detection of fluoride. Solutions of 1.1 either in DCM or in aqueous media at physiological pH show the excellent selectivity towards fluoride to afford color formation while solutions of other interfering anions remain colorless.
