Structural Insights Into The Cl-Par-4 Protein: Ionic Requirements, Conformational Transitions, And Interaction With Cisplatin, Krishna Kumar Raut

Chemistry & Biochemistry Theses & Dissertations

Cancer continues to be the leading global cause of death, with challenges in early diagnosis, drug resistance, non-specific drug targeting, and cancer recurrence and metastasis posing formidable obstacles in cancer therapy. In this context, Prostate Apoptosis Response-4 (Par-4), a pro-apoptotic tumor suppressor protein, emerged as a promising therapeutic target due to its ability to selectively induce apoptosis in cancer cells, thereby minimizing the drug-associated adverse effects. However, a comprehensive understanding of the structural features of Par-4, specifically the caspase-cleaved fragment (cl-Par-4), is crucial for therapeutic advancements.

This dissertation investigated the effects of various ions, both monovalent and divalent, on the …

Student and Faculty Publications

BACKGROUND AND PURPOSE: Automatic review of breast plan quality for clinical trials is time-consuming and has some unique challenges due to the lack of target contours for some planning techniques. We propose using an auto-contouring model and statistical process control to independently assess planning consistency in retrospective data from a breast radiotherapy clinical trial.

MATERIALS AND METHODS: A deep learning auto-contouring model was created and tested quantitatively and qualitatively on 104 post-lumpectomy patients' computed tomography images (nnUNet; train/test: 80/20). The auto-contouring model was then applied to 127 patients enrolled in a clinical trial. Statistical process control was used to assess …

Precise Method To Identify Kinase Drug Targets In Complex Diseases: The First Step Towards Sustainable And Effective Treatment, Hasbanny Irisson, Marzieh Ayati Sep 2023

Precise Method To Identify Kinase Drug Targets In Complex Diseases: The First Step Towards Sustainable And Effective Treatment, Hasbanny Irisson, Marzieh Ayati

Research Symposium

Background: Kinases are enzymes that have proven to be important drug targets due to their role in critical biological mechanisms such as phosphorylation. Phosphorylation happens when a kinase catalyzes the transfer of a phosphate group to a protein in a phosphorylated site, which then becomes known as the substrate of the kinase. Any dysregulation of protein phosphorylation causes a wide range of complex diseases including cancer. Thus, discovering the links between kinases and their substrates (i.e. predicting kinase-substrate associations (KSAs)) is crucial in developing effective and sustainable treatments. Presently, less than 5% of phosphorylated sites have an associated kinase, and …

Comparative Study Of Guanidine-, Acetamidine- And Urea-Based Chloroaluminate Electrolytes For An Aluminum Battery, Iwan Sumarlan, Anand Kunverji, Anthony J Lucio, A Robert Hillman, Karl S Ryder

Student and Faculty Publications

Aluminum-based batteries are a promising alternative to lithium-ion as they are considered to be low-cost and more friendly to the environment. In addition, aluminum is abundant and evenly distributed across the globe. Many studies and Al battery prototypes use imidazolium chloroaluminate electrolytes because of their good rheological and electrochemical performance. However, these electrolytes are very expensive, and so cost is a barrier to industrial scale-up. A urea-based electrolyte, AlCl3:Urea, has been proposed as an alternative, but its performance is relatively poor because of its high viscosity and low conductivity. This type of electrolyte has become known as an ionic liquid …

Modeling Nonsegmented Negative-Strand Rna Virus (Nnsv) Transcription With Ejective Polymerase Collisions And Biased Diffusion, Felipe-Andres Piedra Sep 2023

Modeling Nonsegmented Negative-Strand Rna Virus (Nnsv) Transcription With Ejective Polymerase Collisions And Biased Diffusion, Felipe-Andres Piedra

Research Symposium

Background: The textbook model of NNSV transcription predicts a gene expression gradient. However, multiple studies show non-gradient gene expression patterns or data inconsistent with a simple gradient. Regarding the latter, several studies show a dramatic decrease in gene expression over the last two genes of the respiratory syncytial virus (RSV) genome (a highly studied NNSV). The textbook model cannot explain these phenomena.

Methods: Computational models of RSV and vesicular stomatitis virus (VSV – another highly studied NNSV) transcription were written in the Python programming language using the Scientific Python Development Environment. The model code is freely available on GitHub: …

Self-Organization For Community Resilience In An Invisible Agricultural Community, Anna Erwin, Chelsea A. Silva, Zhao Ma

School of Earth, Environmental, and Marine Sciences Faculty Publications and Presentations

This study investigates how self-organizing efforts by residents of informal settlements, primarily migrant and informal farmworkers, shape community resilience in Majes, a water-scarce irrigation district in the Atacama Desert of Peru. We collected 45 semi-structured interviews with residents and authorities in Majes and analyzed findings through a framework of self-organizing. Analyses revealed that self-organizing by residents of informal settlements incorporated the three components of White’s theory of Community Agency and Community Resilience, which contends that marginalized communities increase resilience by fostering a commons praxis, practicing a prefigurative politics, and developing opportunities for economic autonomy. We also found …

Structural Control Of Metallothionein Metallation And Oxidation, Natalie C. Korkola Sep 2023

Structural Control Of Metallothionein Metallation And Oxidation, Natalie C. Korkola

Electronic Thesis and Dissertation Repository

Metallothioneins (MT) are a family of cysteine-rich metal-binding intrinsically disordered proteins that are ubiquitous across life. The proposed functions of MTs are to maintain Zn(II) homeostasis, participate in heavy metal detoxification, and protect against oxidative stress by binding to a variety of metals in a range of stoichiometries. However, due to their disordered nature and ability to form multiple metallated species, these diverse structures are not well characterized. Electrospray ionization mass spectrometry (ESI-MS) is a technique that allows quantification of heterogeneous metallation states through analysis of the speciation distributions. In this thesis, ESI-MS in combination with other spectroscopic techniques is …

Prioritization Of Scientific Sources Of Water Information: The Effect Knowledge, Beliefs, And Political Identity, Sadie Hundemer

Journal of Applied Communications

Scientists are reported to be more trusted than other information sources; yet, on essential water facts, people sometimes reject what they perceive water scientists to believe in favor of other belief determinants. This study examines the factors that affect the difference in people's stated willingness to reconsider their water beliefs in response to information provided by scientists relative to information provided by other sources. Regression analysis of responses provided by 806 Florida and Georgia residents found water science knowledge to be a consistently strong influencer of the gap in reliance on scientific information providers relative to other sources. Synthesis, Characterization, Antibacterial Activity, And Potential Water Filter Application Of Copper Oxide/Zeolite Composite, Elfa Aida Kamila, Zaenal Abidin, Irma Isnafia Arief, Trivadila Trivadila

Makara Journal of Science

Detection of E. coli bacteria in water samples can indicate the presence of other bacterial contamination in feces. Bacterial contamination can be treated with antibiotics, but excessive use of antibiotics leads to the emergence of multiresistant bacteria. Therefore, alternative antibacterial agents must be explored. Copper ion/zeolite composite has been widely studied as an antibacterial agent. However, the released copper ions accumulate in water and are toxic to humans; this can be prevented by converting copper ions into copper oxides (CuO and Cu2O). In this study, copper oxide/zeolite composite is synthesized and characterized using the heating method, followed by …

The Influence Of Polystyrene Microplastics On Juvenile Steelhead Trout (Oncorhynchus Mykiss), Kaitlyn Marie Baker Sep 2023

The Influence Of Polystyrene Microplastics On Juvenile Steelhead Trout (Oncorhynchus Mykiss), Kaitlyn Marie Baker

Dissertations and Theses

Mass production of plastic within the past decade has led to over 100 billion tons of plastics being added to the world’s oceans through rivers and effluent disposal and decomposition. For marine environments, the sudden and constant growth of microplastics (plastics 1 µm to 5 mm in diameter), is of particular concern to top-predatory fish such as steelhead trout (Oncorhynchus mykiss), who passively or actively uptake microplastics while consuming prey. Previous research has demonstrated clear detrimental impacts of microplastic accumulation in bivalves, seabirds, and zebrafish, including decreased diet, reproduction rates, and metabolism, as well as increased rates of …
