We have analyzed Cluster magnetic field and plasma data during high‐altitude cusp crossing and compared them with high‐resolution MHD simulations. Cluster encountered a diamagnetic cavity (DMC) during northward interplanetary magnetic field (IMF) conditions, and as the IMF rotated southward, the spacecraft reencountered the cavity more at the sunward side of the cusp because the reconnection site had changed location. We found evidence of magnetic reconnection both during northward and southward IMF conditions. The Cluster separation was ∼5000 km, enabling for the first time measurements both inside the DMC and surrounding boundaries that allowed us to construct the structure of the …

Thomson scattering (TS) from electron beams produced in laser-plasma accelerators may generate femtosecond pulses of quasi-monochromatic, multi-MeV photons. Scaling laws suggest that reaching the necessary GeVelectron energy, with a percent-scale energy spread and five-dimensional brightness over 10^16 A/m^2, requires acceleration in centimeter-length, tenuous plasmas (n ~ 10^17 cm^-#3;3), with petawatt-class lasers. Ultrahigh per-pulse power mandates single-shot operation, frustrating applications dependent on dosage. To generate high-quality near-GeV beams at a manageable average power (thus affording kHz repetition rate), we propose acceleration in a cavity of electron density, driven with an incoherent stack of sub-Joule laser pulses through a millimeter-length, dense plasma …

A computer program tailored for EMP damage analysis of solid-state circuitry has been developed by modifying the existing TRAC network analysis program. Modification of the TRAC diode and transistor models to include breakdown parameters and the addition of a semiconductor device parameter library have greatly simplified the analyst's task. An added feature is a subroutine that automatically calculates the amplitude and duration of transient power dissipated in electronic circuit components.

We present a local mesoscale model of the magnetospheric cusp region with high resolution (up to 300 km). We discuss the construction and implementation of the initial configuration and give a detailed description of the numerical simulation. An overview of simulation results for the case of strongly northward interplanetary magnetic field (IMF) is then presented and compared with data from Cluster 2 spacecraft from 14 February 2003. We present results from mesoscale simulations of the magnetospheric cusp region for both strongly northward and strongly southward interplanetary magnetic field (IMF). Simulation results indicate an extended region of depressed magnetic field and strongly enhanced plasma B which exhibits a strong dependence on IMF orientation. These structures correspond to the Cusp Diamagnetic Cavities (CDC’s). The typical features of these CDC’s are generally well reproduced by the simulation. The inner boundaries between the CDC and the magnetosphere are gradual transitions which form a clear funnel shape, regardless of IMF orientation. We have estimated the ionospheric location, area, and travel time of quasi-periodic oscillations originating from the magnetospheric flanks. This was accomplished by utilizing global and local MHD models and Tsyganenko semi-empirical magnetic field model on multiple published and four new cases believed to be caused by the Kelvin– Helmholtz Instability. Finally, we used auroral, magnetometer, and radar instruments to observe the ionospheric signatures. The ionospheric magnetic latitude determined using global MHD and Tsyganenko models ranged from 58.3–80.2 degrees in the Northern Hemisphere and −59.6 degrees to −83.4 degrees in the Southern Hemisphere. Global hybrid (electron fluid, kinetic ions) and fully kinetic simulations of the magnetosphere have been used to show surprising interconnection between shocks, turbulence and magnetic reconnection. In particular collisionless shocks with their reflected ions that can get upstream before retransmission can generate previously unforeseen phenomena in the post shocked flows: (i) formation of reconnecting current sheets and magnetic islands with sizes up to tens of ion inertial length. (ii) Generation of large scale low frequency electromagnetic waves that are compressed and amplified as they cross the shock. New capabilities for studying the Sun allow us to image for the first time the magnetic Kelvin–Helmholtz (KH) instability developing at the surface of a fast coronal mass ejecta (CME) less than 150 Mm above the solar surface. We conduct a detailed observational investigation of this phenomenon, observed off the east solar limb on 2010 November 3, in the EUV with SDO/AIA. In conjunction with STEREO-B/EUVI, we derive the CME source surface position. We ascertain the timing and early evolution of the CME outflow leading to the instability onset. We have performed 13 three-dimensional global magnetohydrodynamic (MHD) simulations of the magnetosheath plasma and magnetic field properties for Parker spiral (PS) and ortho-Parker spiral interplanetary magnetic field (IMF) orientations corresponding to a wide range of solar wind plasma conditions. To study the growth of the Kelvin-Helmholtz instability on the dawn and dusk flank magnetopause, we have performed 26 local two-dimensional MHD simulations, with the initial conditions taken from global simulations on both sides of the velocity shear layer at the dawn-dusk terminator. On 3 July 2001, the four Cluster satellites traversed along the dawnside magnetospheric flank and observed large variations in all plasma parameters. The estimated magnetopause boundary normals were oscillating in the z-direction and the normal component of the magnetic field showed systematic ~2–3 min bipolar variations for 1 h when the IMF had a small positive bz-component and a Parker-spiral orientation in the x, y-plane. Brief ~33 s intervals with excellent deHoffman Teller frames were observed satisfying the Wal´en relation. The recent launch of the equatorial spacecraft of the Double Star mission, TC-1, has provided an unprecedented opportunity to monitor the southern hemisphere dayside magnetopause boundary layer in conjunction with northern hemisphere observations by the quartet of Cluster spacecraft. We present first results of one such situation where, on 6 April 2004, both Cluster and the Double Star TC-1 spacecraft were on outbound transits through the dawnside magnetosphere. Previous works have emphasized the significant influence of the solar wind Alfvén Mach number (MA) on magnetospheric dynamics. Here we report statistical, observational results that pertain to changes in the magnetosheath flow distribution and magnetopause shape as a function of solar wind MA and interplanetary magnetic field (IMF) clock angle orientation. We use all Cluster 1 data in the magnetosheath during the period 2001–2010, using an appropriate spatial superposition procedure, to produce magnetosheath flow distributions as a function of location in themagnetosheath relative to the IMF and other parameters. Background

Buprenorphine is a semi-synthetic opioid used for the treatment of opioid dependence. Opioid use, including buprenorphine, has been increasing in recent years, in the general population and in pregnant women. Consequently, there has been a rise in frequency of neonatal abstinence syndrome (NAS), associated with buprenorphine use during pregnancy. The purpose of this study was to investigate correlations between buprenorphine and buprenorphine-metabolite concentrations in cord blood and onset of NAS in buprenorphine exposed newborns.

Like many hemoglobins, the structure of the dimeric hemoglobin from the clam Scapharca inaequivalvis is a “closed bottle” since there is no direct tunnel from the oxygen binding site on the heme to the solvent. The proximal histidine faces the dimer interface, which consists of the E and F helicies. This is significantly different from tetrameric vertebrate hemoglobins and brings the heme groups near the subunit interface. The subunit interface is also characterized by an immobile, hydrogen-bonded network of water molecules. Large conformational transitions play an essential role in the function of many proteins, but experiments do not provide the atomic details of the path followed in going from one end structure to the other. For the hemoglobin tetramer, the transition path between the unliganded (T) and tetraoxygenated (R) structures is not known, which limits our understanding of the cooperative mechanism in this classic allosteric system, where both tertiary and quaternary changes are involved. The advent of language implementation tools such as PyPy and Truffle/Graal have reinvigorated and broadened interest in topics related to automatic compiler generation and optimization. Given this broader interest, we revisit the Futamura Projections using a novel diagram scheme. Through these diagrams we emphasize the recurring patterns in the Futamura Projections while addressing their complexity and abstract nature. We anticipate that this approach will improve the accessibility of the Futamura Projections and help foster analysis of those new tools through the lens of partial evaluation.

In this review, we summarize the recent literature on dapE-encoded N-succinyl-l,l-diaminopimelic acid desuccinylase (DapE) enzymes, with an emphasis on structure–function studies that provide insight into the catalytic mechanism. Crystallographic data have also provided insight into residues that might be involved in substrate and hence inhibitor recognition and binding. These data have led to the design and synthesis of several new DapE inhibitors, which are described along with what is known about how inhibitors interact with the active site of DapE enzymes, including the efficacy of a moderately strong DapE inhibitor.

The H355A, H355K, H80A, and H80K mutant enzymes of the argE-encoded N-acetyl-L-ornithine deacetylase (ArgE) from Escherichia coli were prepared, however, only the H355A enzyme was found to be soluble. Kinetic analysis of the Co(II)-loaded H355A exhibited activity levels that were 380-fold less than Co(II)-loaded WT ArgE. Electronic absorption spectra of Co(II)-loaded H355A-ArgE indicate that the bound Co(II) ion resides in a distorted, five-coordinate environment and Isothermal Titration Calorimetry (ITC) data for Zn(II) binding to the H355A enzyme provided a dissociation constant (Kd) of 39 μM. The emergence of antibiotic-resistant bacterial strains underscores the importance of identifying new drug targets and developing new antimicrobial compounds. Lysine and meso-diaminopimelic acid are essential for protein production and bacterial peptidoglycan cell wall remodeling and are synthesized in bacteria by enzymes encoded within dap operon. The dapE-encoded N-succinyl-l,l-diaminopimelic acid desuccinylase (DapE) is a critical bacterial enzyme for the construction of the bacterial cell wall. A screen biased toward compounds containing zinc-binding groups (ZBG’s) including thiols, carboxylic acids, boronic acids, phosphonates and hydroxamates has delivered a number of micromolar inhibitors of DapE from Haemophilus influenzae, including the low micromolar inhibitor l-captopril (IC50 = 3.3 μM, Ki = 1.8 μM). In vitro antimicrobial activity was demonstrated for l-captopril against Escherichia coli.

A novel apex-modified cyclotriveratrylene (CTV) derivative with an attached thiolane-containing lipoic acid linker was directly patterned onto gold substrates via dip-pen nanolithography (DPN). The addition of a dithiolane-containing linker to the apex of CTV provides a molecule that can adhere to a gold surface with its bowl-shaped cavity directed away from the surface, thereby providing a surface-bound CTV host that can be used for the directed assembly of guest molecules. Subsequent exposure of these CTV microarrays to C60 in toluene resulted in the directed assembly of predesigned, spatially controlled, high-density microarrays of C60.
