Cloning, Overexpression, Purification And Characterization On N-Terminal Metal Binding Domains 5 And 6 Of Wilsons Disease Protein, David Achila
Masters Theses
Wilsons disease is an autosomal recessive disorder of copper metabolism which results from mutations in the gene encoding for Wilson disease protein (WDP). The cytosolic N-terminus of WDP comprises six metal binding domains each containing a conserved metal binding motif, GMXCXXC. The contribution of these metal binding domains to copper homeostasis is poorly understood. We have cloned, expressed and purified residues 485 to 633 corresponding to N-terminal metal-binding domains 5 and 6 of WDP (WD5-6). Two of the 3 disease causing mutations in the N-terminal metal binding domains of WDP occur one in each of these domains and the two …

Online Support For Multivariate Integration, Shujun Li
Dissertations
In this dissertation, we present an online problem solving environment for multivariate numerical integration. To build an environment for users with various backgrounds, we address issues of Web interface support for end-users, remote application programming interfaces for distributed system programmers, visualization of multivariate integrand functions for problem analysis, and highly efficient integration methods. Compared to previous implementations, our new load balancing schemes increase the scalability of parallel integration and hence are suitable for large scale computations. A parallel integration service, called PI service, was implemented based on Web services technologies to support both remote integration and remote visualization. The multivariate …

Influence Of Uncorrelated Defects On The Vortex Phase Diagram In Yba2Cu3O7-6 Superconducting System, Lucian M. Undreiu
Influence Of Uncorrelated Defects On The Vortex Phase Diagram In Yba2Cu3O7-6 Superconducting System, Lucian M. Undreiu
Dissertations
The goal of our work is to gain more insight into the vortex dynamics in YBa2Cu307-8 single crystals with point defects induced by 9MeV proton irradiation. The position of the critical points from the first order melting transition line can be shifted in a controlled manner with the induced disorder. In order to accomplish a comparative study, different crystals and irradiation doses were used. The results were correlated using both electrical transport and ac specific heat measurements. Special attention was given to investigating the nature of the vortex state below the lower critical point o …
Structure-Activity Relationship Studies On Thioxolone Compounds As Inhibitors Of Human Carbonic Anhydrase Ii, Jared Nyabuto Orwenyo
Structure-Activity Relationship Studies On Thioxolone Compounds As Inhibitors Of Human Carbonic Anhydrase Ii, Jared Nyabuto Orwenyo
Masters Theses
Carbonic anhydrase II (CA II) is a zinc metalloenzyme that catalyzes the reversible hydration of carbon dioxide. The role of this reaction in physiology has made it a target of many therapeutic approaches. Most CA II inhibitors currently known are sulfonamide-based and this is the only functional group incorporated into rationally designed CA II inhibitors. Thioxolone (6-hydroxy-1, 3-benzoxathiol-2-one), is a non-sulfonamide that has been found to have CA II inhibitory activity.
Thioxolone and its analogs were studied to evaluate the inhibition activity that can be achieved upon modification of the thioxolone molecule. IC50 values and dissociation constants were determined …
Design, Synthesis And Characterization Of Nanosensors For Nerve Gas Agents, Thushara Gunasinghe
Design, Synthesis And Characterization Of Nanosensors For Nerve Gas Agents, Thushara Gunasinghe
Masters Theses
This research is focused on the design synthesis and characterization of nanosensors to detect the nerve gas agents such as sarin at very low concentration levels (parts per billion and lower) which is very important to combat terrorism. Nanosensors constructed by the bottom up approach could provide the low sensitivity levels and high selectivity required for nerve gas detection. The sensing mechanism exploits the inherent properties of nanomaterials and signal amplification by signal transduction. The components of a nanoparticle-fluorescent monomer-nanomolecule-receptor (NMNR) sensor have been designed, synthesized, and characterized successfully. Interaction of the components of NMNR sensor with a model nerve …
Investigating Nickel Partitioning In A Contaminant Aquifer, Suama Ndengu
Investigating Nickel Partitioning In A Contaminant Aquifer, Suama Ndengu
Masters Theses
Improved prediction of the transport and bioavailability of metals in contaminated sediments demands a better quantitative understanding of how metals partition between aqueous solutions and mineral phases. This research examines the partitioning of nickel in a contaminated aquifer from Grand Rapids, Ml. Contaminated aquifer sediments were examined using the five step Tessier et al. (1979) operationally defined sequential extraction procedure to determine the solid phase distribution of nickel in the aquifer sediments. Models used to guide remediation efforts at the contaminated site assume that Ni behavior can be predicted based on Ni adsorption onto ferrihydrite (HFO). Although sequential extraction results …

Some hydrocarbon reservoirs have a documented reducing chimney, extending upward to the surface due to diffusion of hydrocarbon volatiles. This effect reportedly reduces the iron in the soil to magnetite and precipitates fine-grained pyrite if there is sufficient sulfur in the system. This study was conducted to test the detectability of the hypothetical chimney, using electrical geophysics in an environment of relatively recent glacial cover, known depth to an underlying petroleum reservoir, and known faulting.
The Calvin 28 oil field, in Cass County Michigan, is a middle Devonian domal field where reservoir rocks are draped over the central uplift block …
A Tool Supporting Validation Of Uml Models, Weng Liong Low
A Tool Supporting Validation Of Uml Models, Weng Liong Low
Masters Theses
Software design is an important phase in the computer software development life cycle. The Unified Modeling Language (UML) is a widely used notation to design software models. As a software system increases in size and complexity it is harder for developers to check that their UML models are correct with respect to the UML metamodel. Consistency among different diagrams in a UML model is also important to ensure that there are no design conflicts that will lead to problems later. Incorrect software design will result in loss of productivity, time and money as developers fix design errors. Therefore, to ensure …

Computer products developed to visualize and integrate meteorological products with real-time data can create a virtual reality of the atmosphere. These advanced technologies help educators to improve teaching techniques in the classroom for science and non-science majors and increase the students' interest in learning as well. One of these products, the Integrated Data Viewer (IDV), was recently developed by University Corporation for Atmospheric Research (UCAR) project, Unidata, to visualize and integrate geoscience data. This study compares students' performance in the laboratory sessions using IDV-based and MANUAL-based exercises. Comparison of lab scores with exam scores of questions on the same topic …
Single-Electron Capture Processes In Slow Collisions Of He^2+ Ions With Molecular Targets, Osama Abu-Haija
Single-Electron Capture Processes In Slow Collisions Of He^2+ Ions With Molecular Targets, Osama Abu-Haija
Dissertations
Translational energy-gain spectroscopy (TES) has been used to investigate state-selective dissociative and non-dissociative single-electron capture processes in low-energy collisions of He2+ ions with O2 , H2 O, CO2 , N2 , and NH3 at impact energies between 100 eV and 1600 eV. The measured energy spectra for theHe2+ -O2 and H2 O collision systems show that the dominant exit channel is due to dissociative transfer ionization (DTI), single-electron captureaccompanied by ionization of the molecular target-ion, at the lowest collision energies. As the impact energy is increased non-dissociative single-electron capture (SEC) into the …

Ionic Liquid As Catalysts In The Cycloaddition Of Co2 To Styrene Oxide In Supercritical And Subcritical States, Joy Mwalimu
Masters Theses
Supercritical fluids (SCFs) have fascinated researchers ever since the discovery of a critical temperature more than a century ago.1 Initial studies focused mainly on the physical properties of supercritical phases.2 However, their use as reaction media has sparked mainly during the last five to ten years.2
The coupling of CO2 and Ionic Liquids (ILs) in the synthesis of cyclic carbonates comes from an increasing need for environmentally and toxicologically benign processes for the production of high-value compounds. In my research, CO2 was utilized as both a replacement solvent and a C1 feedstock in its …
Acylpyrazolones: Synthesis, Self-Assembly And Lanthanide Metal Ion Separation, Jun Yang
Acylpyrazolones: Synthesis, Self-Assembly And Lanthanide Metal Ion Separation, Jun Yang
Dissertations
The central hypothesis that nanoscale self-assemblies can provide excellent metal ion recognition has been substantiated by employing acylpyrazolones and trivalent lanthanide metal ions as model systems. Several novel acylpyrazolones and their amphiphilic analogs have been designed, synthesized, and characterized. Their lanthanide metal ion recognition efficacies have been demonstrated through baseline separations of a mixture of light, middle, and heavy lanthanide metal ions by employing them in the aqueous mobile phase of high performance liquid chromatography (HPLC) with octadecylsilanized silica (ODS) as the stationary phase. The complex separation mechanism is influenced by the structures of acylpyrazolone and amphiphilic moieties, and spontaneous …

The nanoparticles were 2.5 ± 0.5 nm and 5.0 ± 0.5 nm diameter depending on the synthetic procedure employed and the amount of Mn doped varied from 0.003 to 0.32% (w/w). The nanoparticles were characterized by UV-Vis absorption, luminescence, EPR, near edge x-ray absorption fine structure (NEXAFS), and x-ray photoelectron spectroscopy (XPS), and x-ray diffraction (XRD), and transmission electron microscopy (TEM). These quantum dots show a strong emission at 590 nm due to the 4T1 -6A1 transition characteristic of Mn …
Rank-Based Methods For Repeated Measures Data Under Exchangeable Errors, John Kloke
Rank-Based Methods For Repeated Measures Data Under Exchangeable Errors, John Kloke
Dissertations
Rank-based estimation methods provide alternatives to least squares. Estimators derived via least squares are generally not robust to aberrant observations.Rank-based methods for linear models generalize traditional Wilcoxon procedures in the simple location models and are robust.
In the usual linear model it is assumed that the errors are independent. In the case of repeated measures data several observations are taken on each experimental unit. In the case of longitudinal data the measures are taken on the same subject over time. As such an independence assumption does not seem valid. A common solution to this is to make an assumption on …

This research was aimed at: 1) quantifying redox-sensitive parameters in order to study redox stratification of the upper freshwater sediments at Asylum Lake and Kleinstuck Marsh, 2) quantifying the speciation of metals (e.g., Pb, Mn, Fe, Cr, Cu, Zn, Co and Cd), particularly with respect to their association with Fe and Mn (hydr)oxide phases, and 3) assessing changes in redox stratification and metal speciation as a function of season in the sediments. Both the solid phase and sediment pore waters were collected to a depth of ~50cm. Pore waters were analyzed for redox-sensitive species and the sediment solid phase was …
Structure Preserving Algorithms For Computing The Symplectic Singular Value Decom Position, Archara Chaiyakarn
Structure Preserving Algorithms For Computing The Symplectic Singular Value Decom Position, Archara Chaiyakarn
Dissertations
In this thesis we develop two types of structure preserving Jacobi algorithms for com puting the symplectic singular value decomposition of real symplectic matrices and complex symplectic matrices. Unlike general purpose algorithms, these algorithms produce symplectic structure in all factors of the singular value decomposition.
Our first algorithm uses the relation between the singular value decomposition and the polar decomposition to reduce the problem of finding the symplectic singular value decomposition to th a t of calculating the structured spectral decomposition of a doubly structured m atrix. A Jacobi-like m ethod is developed to compute this doubly structured spectral decomposition. …

Stratification And Domination In Graphs And Digraphs, Ralucca M. Gera
Dissertations
In this thesis we combine the idea of stratification with the one of domination in graphs and digraphs, respectively.
A graph is 2-stratified if its vertex set is partitioned into two classes, where the vertices in one class are colored red and those in the other class are colored blue. Let F be a 2-stratified graph rooted at some blue vertex v . An F -coloring of a graph G is a red-blue coloring of the vertices of G in which every blue vertexu belongs to a copy of F rooted at u . The F -domination number γ …

An Investigation Of The Interlobate Area West Of Three Rivers, Michigan And Its Relationship To The Late Wisconsinan Glaciation Of Southwest Michigan, Scott J. Kendzierski
Masters Theses
A detailed investigation of selected mineralogical and textural characteristics of glacial deposits was conducted in order to explore the complex relationships between the Lake Michigan and Saginaw Lobe ice-margins and associated depositional regimes during the deglaciation of southwest Michigan. Various analytical methodologies were applied to the characterization of glacial materials, glacial and post-glacial environments in order to infer a chronology of lobate activity.
The Sturgis Moraine, located in St. Joseph County, Michigan, is a major recessional moraine owing to the processes of both the Lake Michigan and Saginaw Lobes. Analysis of geologic materials and examination of subsurface data along with …
Glacial Stratigraphy And Surficial Geology Of The Decatur, Lawrence, And Paw Paw U.S.G.S.7.5 Minute Quadrangles In Van Buren County, Michigan, Brian Bird
Masters Theses
The modern day landscape of Van Buren County Michigan was created by the advance and retreat of glaciers from about 13,000 to 20,000 years ago. The Lake Michigan Lobe of the Laurentide Ice Sheet sculpted southwest Michigan most recently. The sediments deposited by the glacier include diamicton, outwash sediments, and lacustrine deposits. The build up of sediment by the glaciers led to the construction of particular features such as moraines, till plains, and outwash fans. The huge amount of meltwater from the glaciers led to the erosion of channels throughout the area. In the area of this investigation the Ganges …

Synthetic And Structural Studies Of Novel 1, 10-Phenanthroline Derivatives, Eunjoo Kim
Masters Theses
Asymmetric catalysis can provide a general approach to the preparation of enantiomerically enriched compounds in organic synthesis. Very often these asymmetric catalysts consist of a chiral ligand-metal complex. Because of the growing demand to develop enantioselective reactions in organic chemistry one focus of this area of research is the design and characterization of new chiral ligands for asymmetric synthesis.
The focus of my research is in the design, synthesis, and characterization of novel 1, 10-phenanthroline derivatives that could be used as chiral ligands in enantioselective reactions. Central to these studies have been the effect of substituents on the B-ring of …
Copper Exchange Studies Between The First And Fourth N-Terminal Metal-Binding Domains Of Wilson Disease Atpase, Jennifer Bunce
Copper Exchange Studies Between The First And Fourth N-Terminal Metal-Binding Domains Of Wilson Disease Atpase, Jennifer Bunce
Masters Theses
The human copper-transporting ATPase Wilson's disease protein was one of the first proteins shown to be actively involved in cellular copper homeostasis. The protein includes six N-terminal metal binding domains. The individual capabilities of the domains to bind and transfer copper are not yet fully understood. Each metal-binding domain is about 72 amino acid residues long and contains the conserved binding motif GMTCXXC, of which the cysteines are involved in the copper coordination. Studies have shown that the protein binds six copper atoms suggesting the potential involvement of each metal-binding domain. Separate functional roles for the latter domains have been …
Rhizopine Synthesis – Potential Mediators For Nitrogen Fixation In Legume Plants, Venkat Reddy Guduru
Rhizopine Synthesis – Potential Mediators For Nitrogen Fixation In Legume Plants, Venkat Reddy Guduru
Masters Theses
This research project focused on novel and efficient synthetic approaches of inosamine derivatives (rhizopines). The preparation of specific rhizopines would help elucidate the microbial pathway of rhizopine catabolism and degradation, and determine the role of corresponding genes during nitrogen fixation. We have investigated regio - and stereoselective transformations of myo-inositol derivative 28 utilizing an organostannane as key intermediate towards rhizopine 1a. The synthesis involved several protection and deprotection strategies to control competing reactivities of six hydroxyl groups of myo-inositol. Rhizopine 1a was prepared efficiently as a single isomer in 8 steps and 13% yield by controlling reactions …
Effect Of Antioxidants On Cadmium Induced Oxidative Stress And Signal Transduction, Deepti Goel
Effect Of Antioxidants On Cadmium Induced Oxidative Stress And Signal Transduction, Deepti Goel
Masters Theses
A study was done to establish if reactive oxygen species (ROS) participate in the signal transduction pathway through which cadmium induces gene transcription. Cadmium has become a concern in the health care community in the past few years as evidence suggests that cadmium can induce the development of tumors, influence cell signaling and cause damage to organs, tissue, cells and DNA. The mechanism by which cadmium exerts its action on cellular processes or induces toxicity is however unclear although there is evidence to suggest that ROS play a role. Our laboratory is interested in understanding the mechanisms of differentially regulated …
Global Optimality Conditions In Mathematical Programming And Optimal Control, Pariwat Pacheenburawaa
Global Optimality Conditions In Mathematical Programming And Optimal Control, Pariwat Pacheenburawaa
Dissertations
We derive new first-order necessary and sufficient optimality conditions characterizing global minimizers in mathematical programming and optimal controlproblems. These conditions are based on level sets of an objective functional and they do not assume special structure of a problem (convexity, linearity, etc.). For a mathematical programming problem of minimization of a smooth functional on some compact convex set with equality nonlinear constraints, we derive first-order optimality conditions in the form of a generalized Lagrange multiplier rule. This rule should hold for any point from the level set of the objective functional corresponding to a global minimizer. We demonstrate that these …

Synthesis And Characterization Of A Recombinant Thrombin Inhibitor, Joel Lwande
Synthesis And Characterization Of A Recombinant Thrombin Inhibitor, Joel Lwande
Masters Theses
The final step of the blood coagulation cascade is catalyzed by thrombin, a serine protease. Like most enzyme activities, thrombin activity encounters inhibition from both direct and indirect inhibitors. A new thrombin inhibitor peptide named theromin was recently isolated from the leech Theromyzon tessulatum, and it is the most potent thrombin inhibitor isolated to date. Theromin is made up of 67 amino acid residues, out of which 16 are cysteines engaged in 8-disulfide bridges.
We synthesized the theromin gene and expressed the protein in Eschericia coli. We purified the recombinant protein and carried out thrombin inhibition assays that …

Chapter I
Introduction
Southern Michigan's geomorphology is a testament to the glacial processes that buried and sculpted the landscape during the last glaciation. Many glacial landforms in southwestern Michigan, including the Kalamazoo River Valley, provide evidence that aids in understanding the chronology, ice dynamics and glacial processes that occurred in Michigan during the last ice age.
As the climate began to warm about 20,000 yr B.P. (Larson and Schaetzl, 2001) the extensive Laurentide Ice Sheet began to recede from its maximum southern position reached during the Late Wisconsinan (Figure 1). It was at this point that modem surficial drainage systems …
