The main object in PCR is a primer, a short single strand of DNA, about 18-25 bases long, that serves as the starting point of DNA synthesis. Primers are essential for DNA replication because the enzymes that catalyze this process, DNA polymerases, can only add new nucleotides to an existing strand of DNA. Global climate models (GCM) are sophisticated numerical models used to make long term climate projections. However, the resolution of their output is too coarse for climate change related local impact studies on urban regional scales. Downscaling efforts are taken to address this and increase GCM projection resolution. This thesis develops several strategies for calculating ruin-related quantities for a variety of extended risk models. We focus on the Sparre-Andersen risk model, also known as the renewal risk model. The idea of arbitrary distribution for the waiting time between claim payments arose in the 1950’s from the collective risk theory, and received many extensions and modifications in recent years. Our goal is to tackle model assumptions that are either too relaxed for traditional methods to apply, or so complicated that elaborate algebraic tools are needed to obtain explicit solutions.

This thesis focuses on the synthesis of a variety of heterocycles and carbocycles such as pyrrolidines, dihydrofurans, dihydropyrroles, cyclopentenes, and 2,5-dihydrooxepines. Traditional synthetic methods are discussed first, followed by the manipulation of donor-acceptor cyclopropanes towards the above-mentioned compounds. This leads to a discussion about an important pyrrolidine-containing natural product known as kainic acid. Kainic acid has been studied in the neuroscience field for its biological properties which have led to a better understanding of common human neurological disorders like epilepsy, Huntington’s disease and the after-effects of strokes.

The work disclosed in this dissertation outlines novel reactions involving indoles and their applications towards the total synthesis of natural products, tronocarpine and dippinine B.

Showcased in Chapter 2 is a novel mode of activation for donor-acceptor cyclopropanes via an external hydrogen bond. The hydrogen bond increases the cyclopropane’s electrophilicity permitting indole nucleophiles to open the ring. The result is 3-postion functionalized indoles.

An external hydrogen-bond donor, HFIP (1,1,1,3,3,3-hexafluoroisopropanol), is used as a solvent to provide the medium necessary for favourable hydrogen-bond interaction with donor-acceptor cyclopropanes. Photopolymer networks with phosphonium cation, alkyl phosphine and olefin functionality were designed, synthesized and functionalized with metals by metathesis, coordination and hydrometallation reactions, respectively. The materials were strategically designed so that the metal functionalization step could be monitored and quantified. In some cases, this involved characterization by IR, NMR, or X-ray spectroscopic techniques, or by comparison to molecular analogues. It was found that by using a bi-functional photopolymer network, the material could be bi-metallized by orthogonal mechanisms. There is a collective prediction among ecologists that climate change will enhance phytoplankton biomass in temperate lakes. Yet there is noteworthy variation in the structure and regulating functions of lakes to make this statement challengeable and, perhaps, inaccurate. To generate a common understanding on the trophic transition of lakes, I examined the interactive effects of climate change and landscape properties on phytoplankton biomass in 12,644 lakes located in relatively intact forested landscapes. Chlorophyll-a (Chl-a) concentration was used as a proxy for phytoplankton biomass. It is a non-intuitive but well-established fact that the first and higher vertical ionization energies (VIE) of any N-electron system are encoded in the system's ground-state electronic wave function. This makes it possible to compute VIEs of any atom or molecule from its ground-state wave function directly, without performing calculations on the (N-1)-electron states. By studying achondrite meteorites, we can bring further geochemical and chronological constraints on the source and composition of materials and their evolution throughout the protoplanetary disk. The growth hormone secretagogue receptor-1a (GHS-R1a) is expressed in many human tissues, most notably the hypothalamus, and causes an increase in appetite upon activation by its endogenous ligand, ghrelin. GHS-R1a is differentially expressed in malignant compared to benign tumours. Therefore, there is an interest in developing GHS-R1a-targeted peptides as novel drugs to modulate signaling for diseases such as cancer cachexia and obesity and to image the receptor for disease diagnosis and following progression.

Weathering processes on Ni-rich ultramafic rocks in tropical areas produce laterites that become exploitable for Ni mining. To better understand these processes, samples were collected stratigraphically with water samples from several Ni mines in the Philippines and studied for Ni and Fe isotope compositions. This study found that Ni isotope fractionation takes place during the formation of Ni-enriched minerals (goethite), when light Ni isotopes are preferentially incorporated into new minerals formed, leaving heavy Ni isotopes in groundwater. One of the first steps of methylmercury (MeHg) bioaccumulation, uptake by phytoplankton, is regulated by dissolved organic matter (DOM). Methylmercury binds strongly with DOM, especially high-molecular weight (HMW) compounds containing reduced sulphur; this binding decreases MeHg bioavailability. Climate-induced changes were measured at the landscape level using ArcGIS; land-cover changes between 2005-2010 were assessed around lakes in Mackenzie Valley, NWT. The observed changes, a 10% increase in forest-classified area and up to 15% decrease in wetland area, will likely increase delivery of HMW DOM and MeHg to lakes. The serviceability limit state design approaches in particular are based on simple isotropic elastic half-space analyses that ignore coupling between loading directions, and soil non-linearity and elastic anisotropy. Regression analyses in epidemiological and medical research typically begin with a model selection process, followed by inference assuming the selected model has generated the data at hand. It is well-known that this two-step procedure can yield biased estimates and invalid confidence intervals for model coefficients due to the uncertainty associated with the model selection. To account for this uncertainty, multiple models may be selected as a basis for inference. This method, commonly referred to as model-averaging, is increasingly becoming a viable approach in practice.
