The Effects Of Dissolved And Suspended Solids On Freshwater Meiofauna, Jessica Cline

Meiofauna include small-sized animals (< 1mm) distributed in all aquatic ecosystems on Earth, where they play fundamental trophic and ecological roles. The biodiversity of marine meiofauna and its links with anthropogenic activities is routinely investigated, however, freshwater communities are less known. This is particularly true for the Southeastern United States, which is greatly investigated and elected a hotspot of biodiversity for larger species but very little is known about the meiofauna. The purpose of this research is to reveal the biodiversity of meiofauna from the Tennessee River and test for potential correlations with anthropogenic activities. As a proxy for pollution, dissolved and suspended solids were considered in this study. The research hypothesis is that meiofaunal biodiversity would be affected by possible changes of dissolved and suspended solids in the water column. Possible mechanisms causing biodiversity shifts could be ascribed to osmotic stresses of animals to cope with variation in dissolved solids or, more indirectly, because different sunlight penetration caused by suspended solids would affect primary production. To test the hypothesis, water samples were collected from nine stations located along the Tennessee River in Hamilton County. Each station was visited three times, and, during each visit, environmental parameters (including dissolved and suspended solids) were measured. Meiofauna biodiversity (estimated as richness, community composition, and phylogenetic diversity) was revealed using a metagenomic approach. Statistical analyses were applied to test for possible correlations between the biodiversity estimates and the measured environmental parameters. Results show a high biodiversity of meiofauna with more than 200 amplicon-sequence variants distributed across 10 metazoan phyla. Environmental conditions are highly variable among stations and statistical analyses show that while both dissolved solids (TDS) and turbidity (suspended solids, NTU) did not significantly affect meiofauna biodiversity in the collected samples, various other water, and sediment metrics were found to be significant predictors of meiofauna biodiversity. In conclusion, the results of this project not only reveal for the first time the meiofauna biodiversity from the Tennessee River, but also suggest that meiofauna could be used as bioindicators for several anthropogenic activities in freshwater ecosystems.

Luminescent transition metal complexes are of interest for biological imaging applications due to their unique photophysical properties including large Stokes shift, long fluorescence lifetimes, and tunable emission. Altering ligand structure is a useful way to manipulate the charge transfer (CT) and mixed character states that give these complexes their photophysical properties. A series of organometallic complexes containing ligands with externally facing nitrogens has been prepared and synthesized. Complexes of the form Pt (II)(tbbpy)(C2-py), Re(CO)3(N^N), Ir(C^N)2(N^N), and Ir(C^N)2(N^O) have been studied for their potential application as biological imaging agents. Conjugated polymers offer the potential for the development of robust, low-cost electronics, but achieving high mechanical deformability and high charge transport simultaneously in polymer semiconductors remains a significant challenge. In this work, blends of conjugated polymers were investigated to elucidate the influence of compatible conjugated blends on polymer morphology, mechanics, and electrical properties (using a partially conjugated polymer as the soft matrix and a fully conjugated polymer as the electrically active component). Sea breezes (SB) occur frequently from May through October along the U.S. Gulf of Mexico Coast. One reason why SBs interest forecasters is their ability to spawn summertime thunderstorms, which can cause flash flooding, lightning, and hail. Coordination polymers are repeating units of a coordination complex that have applications as catalysts, conductors, and magnets. In this work, silver (I) was the metal of choice because it behaves similarly to copper (I), which has been studied much more extensively and thione ligands were chosen due to their potential for forming coordination polymers. These coordination complexes with silver were synthesized by combining silver nitrate with thioacetamide, N,N'-diphenylthiourea, and 2-mercapto-1-methylimidazole. These complexes were characterized via IR spectroscopy, elemental analysis, and X-ray crystallography. Our laboratory has reported the synthesis and characterization of a broad library of unsymmetrical bidentate bis(phosphino)pyrrole (BPP) ancillary ligands for use in nickel-catalyzed cross-coupling reactions. The electronic and steric properties of nickel complexes bearing these ligands are examined, and our data support the hypothesis that these ligands produce nickel complexes with electron-deficient metal centers. The syntheses of (BPP)Ni(o-tolyl)Cl precatalysts for utilization in nickel-catalyzed Buchwald-Hartwig Amination are described, and preliminary reactivity studies reveal that (BPP)Ni(o-tolyl)Cl precatalysts are effective in promoting C–N cross-coupling reactions. Collision Induced Dissociation (CID) and Ion Mobility Mass Spectrometry (IM-MS) are methods used to understand the structural arrangement and stability of large molecules. In this study, the gold nanocluster Au30(S-tBu)18 was dissociated at varying Trap collision energies, and drift time vs mass spectrometry data was plotted to determine the order of cluster fragmentation. This thesis focuses on two research projects. The first project reported in Chapter 1 involves the synthesis and application of a tetradentate ligand (2-(1,1-di(pyridin-2-yl)ethyl)-1,10-phenanthroline, PhenPY2Me) that is metalated with iron to give a catalyst capable of carbon-hydrogen (C-H) bond oxidation. C-H bond oxidation provides a method that may allow some transformations in organic syntheses to be streamlined. The mononuclear iron complex supported by PhenPY2Me was found to dimerize in the presence of O2 in a non-coordinating solvent. Fast photochemical oxidation of proteins (FPOP) is a useful tool in proteomics because of the ability for modifications to occur on the scale of microseconds which reduces the modifications to tertiary and quaternary structure allowing for more accurate labeling of the protein. Labels for FPOP are generated from various radicals in our experiments which include hydroxyl radicals and trifluoromethyl radicals. Hydroxyl radicals are easily generated by using an excimer laser (KrF laser, 248 nm) or a UV flash lamp (as a part of the Fox™ System) by the photolysis of hydrogen peroxide. Since fractal surfaces appear naturally in other areas, it must be asked if they can produce less drag than a traditional surface and save energy. 55Fe radioactive sources are used for x-ray calibration due to the long half-lives of 2.73 years and the optimal energies of the peaks. The source used in the experiment was generated using Western Michigan University’s Tandem Van de Graff accelerator to produce a 6 MeV proton beam which induced a 55Mn(p, n)55Fe reaction on a 10 μm 55Mn target. The emissions from this target were then measured using a silicon lithium drift x-ray detector to determine the number of instances of deexcitation through kα and kβ modes. Among estuaries, there is a large block of co-dependent and independent variables shaping habitat and environmental conditions within a critical area. Historical data proxies and indexes from seven different stations were assessed to consider temporal and spatial variability of the marine and non-marine transition along the South Carolina Pee Dee River Basin. Historical datasets from the upper reaches of Winyah Bay and lower section of the Pee Dee River basin were compiled along with shorter termed deployments of a new instrument station installed near Georgetown, SC. Quinoxaline and its derivatives have been studied extensively for their relevant biological activity and transition metal selectivity. These compounds are commonly used for their antimicrobial, antifungal, antiparasitic activity, and relevance in the treatment of metabolic diseases [6]. More recently, quinoxaline’s ability to inhibit gram-positive bacterial growth has been found especially in oxygen- containing compounds, yielding promising candidates to prevent cancerous tumor growth [5]. Though plastic polymers such as polyester polyurethanes have many applications from electronics to aircraft coatings, their resistance to natural degradation presents an environmental concern. Therefore, elucidating the mechanism of degradation from microorganisms that were discovered breaking down the coating of a cargo aircraft may offer insights into a method of bioremediation.
