Tropical cyclones (TCs) pose a significant threat to life and property, and exhibit many severe weather hazards as they make landfall, such as storm surge, strong winds, flooding rains, and tornadoes. TC convection is associated with nearly all of these hazards, which can extend hundreds of kilometers inland; thus, understanding the characteristics and organization of convective cells is important to mitigating risk. Observational studies have noted that TC convection tends to organize downshear and that rotating thunderstorms tend to occur in the downshear-right quadrant of the TC. In the field of applied topology, or more specifically, topological data analysis, we usetools such as the persistent homology of a filtered space to give shape to data. Alongside the persistent homology groups of a space is the barcode invariant. In this thesis, we explore the failure of injectivity of the barcode invariant for merge trees, Reeb graphs, circle embeddings in R2 and S2 embeddings in R3 up to different types of equivalence. This thesis describes a chemical method for alternative oligonucleotide purification that is non-chromatographic and gel-free and allows to routinely synthesize and purify long functional RNA strands. The purification of long RNAs is based on the bio-orthogonal inverse electron demand Diels-Alder (IEDDA) chemistry between trans-cyclooctene (TCO) and tetrazine (Tz). Target oligonucleotide strands are selectively tagged with Tz and can be captured and purified from the failure sequences with immobilized TCO. RNA strands are synthesized on solid support through a photolabile linker to avoid the loss of Tz tag. Stochastic model error schemes, such as the stochastic perturbed parameterization tendencies (SPPT) and independent SPPT (iSPPT) schemes, have become an increasingly utilized method to represent model error associated with uncertain subgrid-scale processes in ensemble prediction systems (EPSs). While much of the current literature focuses on how stochastic methods influence ensemble skill, relatively less attention is given to the processes by which these schemes lead to forecast variability. Atmospheric aerosol has profound impacts on air quality, though particles of differing sizes can have profoundly different characteristics which are understated when measurements are reported as a single parameter (typically number concentration or total mass). To address some of these concerns and to better understand the nature of atmospheric aerosol, continuous particle size distributions have been measured at two sites in New York State. Measurement was performed for particle diameters between 7.6nm - 20µm at Pinnacle State Park (Addison, New York; March 2017-April 2018) and Queens College (May 2018-September 2019) at a 5-minute time resolution. Part I: Koskta-Foulkes polynomials are Lusztig's q-analogues of weight multiplicities for irreducible representations of semisimple Lie algebras. It has long been known that these polynomials can be written with all non-negative coefficients. A statistic on semistandard Young tableaux with partition content, called \textit{charge}, was used to give a combinatorial formula exhibiting this fact in type $A$. Defining a charge statistic beyond type $A$ has been a long-standing problem. In this dissertation we investigate the extension of two applications of focusing polycapillary optics. The first is the application of polycapillary optics in radiation therapy. This discussion includes measurements and calculation of dose for focused beam orthovoltage therapy. A system has been designed to investigate whether the polycapillary optics can produce an X-ray beam which can give more accurate dose painting due to the higher dose concentration at the focal spot. Body fluids, such as blood and semen, can provide invaluable information to a criminal investigation. As such, it is imperative that body fluid evidence is properly detected and identified. Our laboratory has developed a methodology to universally detect and identify body fluids using Raman spectroscopy and chemometrics. This methodology was developed under ideal conditions, which are rare in the field. Understanding the performance of parallel and distributed programs remains a focal point in determining how compute systems can be optimized to achieve exascale performance. Lightweight, statistical models allow developers to both characterize and predict performance trade-offs, especially as HPC systems become more heterogeneous with many-core CPUs and GPUs. This thesis presents a lightweight, statistical modeling approach of performance variation which leverages extreme value theory by focusing on the maximum length of distributed workload intervals.
