Chemistry In Art: The Science Of Dye, Madeleine Gray Burland May 2020

Chemistry In Art: The Science Of Dye, Madeleine Gray Burland

Honors Projects

Fabric arts, and the practice of dyeing fabric using various resist techniques, is a tradition that goes back centuries, and is unique among art mediums in its relation to science, as the innovations in dye production have directly affected the art form. The development of synthetic dyes in the 1800’s greatly affected the way fabric is dyed, and subsequently the way clothes were made and consumed. As opposed to dyes made of natural materials, synthetic dyes cam in more colors, were brighter, easier to make in large quantities, and lasted longer since they didn’t fade with repeated washings. The practice …

Approaches Towards The Synthesis Of Ketamine Metabolites, Ann K. Patrick May 2020

Approaches Towards The Synthesis Of Ketamine Metabolites, Ann K. Patrick

Honors Theses

Major Depressive disorder (MDD) plagues society and stands at the forefront of research as MDD affects approximately 16% of the population. Pharmaceutical drugs, including the selective serotonin reuptake inhibitors (SSRIs), have been used for MDD treatment and remain a popular option today. However, current antidepressant treatments have proven to be ineffective in just less than half of the patients. Research continues with the goal to better understand the mechanisms of the pathology of depression and to search for other treatment options. For example, the stress-neurogenesis hypothesis investigates the role of stress and decreased neuroplasticity within MDD.

Supporting the stress-neurogenesis hypothesis, …

Surface-Enhanced Raman Spectroscopy For Cocaine Detection, Kiersten Jo Mead May 2020

Surface-Enhanced Raman Spectroscopy For Cocaine Detection, Kiersten Jo Mead

Chemistry

The detection of drugs of abuse using Raman spectroscopy is of particular interest to forensic researchers at the moment. Raman spectroscopy is highly specific, fast, non-destructive, and can be adapted for in-situ measurements, making it the ideal forensic technique. Researchers working under the guidance of Doctor Igor Lednev have been able to use UV resonance Raman spectroscopy to detect cocaine in oral fluid without the need for sample pretreatment. They were however unable to detect the cocaine at forensically relevant levels. To overcome this limitation, surface-enhanced Raman spectroscopy (SERS) was used in this proof of concept study for the detection …

ZoeJ and Marcoliusprime are mycobacteriophage capable of infecting tuberculosis causing bacteria via the lytic and lysogenic cycles. The lytic cycle is the form of viral replication that results in cell death. Lysogeny involves phage DNA being integrated with the DNA of the host cell and preserved through binary fission. However, the bacteria causing tuberculosis do not die in the lysogenic phase. This research project will involve creating mutants of ZoeJ and Marcoliusprime that will not contain the genes …

Mycobacteriophage Morphology As A Diagnostic For Cluster Assignment, Ethan Dionne

Chemistry & Biochemistry Student Scholarship

Major: Biochemistry

Faculty Mentor: Dr. Kathleen A. Cornely, Chemistry and Biochemistry

Phages are viral bodies that infect bacterial hosts, and have shown promising applications as alternatives to antibiotics for the treatment of bacterial infections. This project examines the morphology of siphoviridae mycobacteriophage, which have long, flexible, non-contractile tails as well as the characteristic head, called a capsid. Using electron microscopy photos of sequenced phages, tail length and capsid diameter were measured and compared to further characterize morphological relationships between genetically distinct phages. The data presented has the potential to work as a diagnostic tool to classify unsequenced phages to genetically …

Breakthroughs In Obtaining Qm/Mm Free Energies, Phillip S. Hudson Apr 2020

Breakthroughs In Obtaining Qm/Mm Free Energies, Phillip S. Hudson

USF Tampa Graduate Theses and Dissertations

The computation of free energy is pivotal to understanding the fundamental nature of chemical phenomena. That is, whether a specific molecular outcome occurs spontaneously or is inherently unfavorable. The need to do this with consistent accuracy begs for the use of quantum mechanical (QM) methods. However, techniques for directly computing free energy differences with QM or mixed QM/MM methods are untenable, as the computational expense is quite exorbitant. At present, the most feasible approach for obtaining QM/MM free energies is employing the so-called indirect cycle, which relies on accurately computing free energy differences between low (e.g., molecular mechanical, MM) and …

Research And Review Of Polymethyl-Methacrylate Copolymers In The Development Of Enhanced Pharmaceutical Delivery Systems, Jake Dyess

Senior Honors Theses

The following thesis shall primarily explore the current research regarding the implementation of modifiable microscopic and nanoscopic polymer particles as novel pharmaceutical delivery systems. Polymethacrylate-based copolymers, such as EudragitTM, are sensitive to alterations in pH levels, becoming increasingly more soluble in response to an increase in pH. The selective solubility and modifiable nature of these polymers allows for greater flexibility of treatment options for patients (Patra et al., 2017). The wide range of functionality of polymethacrylate (PMA) may provide solutions to challenges relating to the current treatment and therapy of certain conditions. Research indicates that PMA based micro- …

Computation Of Atmospheric Concentrations Of Molecular Clusters From Ab Initio Thermochemistry, Tuguldur T. Odbadrakh, Ariel G. Gale, Benjamin T. Ball, Berhane Temelso, George C. Shields

Chemistry Publications

The computational study of the formation and growth of atmospheric aerosols requires an accurate Gibbs free energy surface, which can be obtained from gas phase electronic structure and vibrational frequency calculations. These quantities are valid for those atmospheric clusters whose geometries correspond to a minimum on their potential energy surfaces. The Gibbs free energy of the minimum energy structure can be used to predict atmospheric concentrations of the cluster under a variety of conditions such as temperature and pressure. We present a computationally inexpensive procedure built on a genetic algorithm-based configurational sampling followed by a series of increasingly accurate screening …

Scholarship and Professional Work - LAS

Over 100 metabolic serine hydrolases are present in humans with confirmed functions in metabolism, immune response, and neurotransmission. Among potentially clinically relevant but uncharacterized human serine hydrolases is OVCA2, a serine hydrolase that has been linked with a variety of cancer-related processes. Herein, we developed a heterologous expression system for OVCA2 and determined the comprehensive substrate specificity of OVCA2 against two ester substrate libraries. Based on this analysis, OVCA2 was confirmed as a serine hydrolase with a strong preference for long-chain alkyl ester substrates (>10-carbons) and high selectivity against a variety of short, branched, and substituted esters. Substitutional analysis …

Paradigms For Creating Activities That Integrate Mathematics And Science Topics, Janet Bowers, Kathy Williams, Antoni Luque, Devon Quick, Mary Beisiegel, Jody Sorensen, Joan Kunz, Diane Smith, Lori Kayes

Journal of Mathematics and Science: Collaborative Explorations

Research has shown that undergraduate students benefit from seeing examples of mathematics applied to real-world situations. This article describes three different paradigms for how math and discipline partner faculty worked together to create mathematical activities that illustrate applications of the topics being studied in precalculus and calculus. All three examples are discussed within the framework of PDSA cycles to describe the process by which the teams collaborated to plan, enact, study, and refine their lessons. Findings discuss both the difficulties of creating integrated activities (differences in terms and definitions between math and science faculty, different foregrounding of math versus science …

A Model Experiment To Investigate The Possibility Of Glyphosate Contamination In St. Kitts And Nevis, Winston Fu

Honors Undergraduate Theses

Glyphosate is a broad-spectrum herbicide that is used globally to combat the spread of weeds among crops. Not all countries around the world have been using the herbicide. For example, island countries such as Dominica and St. Kitts and Nevis do not use the herbicide on their crops. However, St. Kitts and Nevis imports food and other agricultural products from around the world, leading to the hypothesis that glyphosate is contaminating the soil through those imported products. The aim of this research project was to determine if glyphosate is present in the island country of St. Kitts and Nevis. A …

Green Chemistry: The Oxidation Of Benzaldehyde Using Atmospheric Oxygen And N-Heterocyclic Carbenes As Catalysts, Alex Brody, John Morgan Jan 2020

Green Chemistry: The Oxidation Of Benzaldehyde Using Atmospheric Oxygen And N-Heterocyclic Carbenes As Catalysts, Alex Brody, John Morgan

Student Research Poster Presentations 2020

Oxidation is a vital process in organic and biochemical reactions. In particular, the oxidation of aromatic aldehydes to carboxylic acids and esters is a vital process used in many different environments. Benzaldehyde is one common subject of this type of reaction, used in the synthesis of benzoic acid and benzoate derivatives. Unfortunately, the industrial synthesis of these compounds uses harmful heavy metal oxidants such as Chromium(VI) and Manganese(VII), which are very harmful to the environment. We proposed to eliminate these pollutants by using atmospheric oxygen and an organic catalyst in a solvent free reaction. This reaction would then be microwaved …

Electrolytic Oxidative Coupling Of Alcohols With Aldehydes To Form Esters, Katherine L. Verboom Jan 2020

Electrolytic Oxidative Coupling Of Alcohols With Aldehydes To Form Esters, Katherine L. Verboom

Honors College Theses

There are copious amounts of naturally occurring and synthetic esters known that have a wide range of applications in industry to produce fine chemicals. Traditional methods of ester synthesis involve a reaction between a carboxylic acid and an alcohol in the presence of a Bronsted acid catalyst. The primary objective of this research project was to carry out the ester synthesis using the electrochemical coupling of aldehydes with alcohols instead of with a Bronsted acid catalyst because the electrochemical coupling would be an all-around better alternative of ester synthesis than the traditional method of using the catalyst. Electrochemistry has long …

Development Of Tetrathiafulvalene Fused N-Heterocyclic Carbene Compounds, William J. Robinson Iii Jan 2020

Development Of Tetrathiafulvalene Fused N-Heterocyclic Carbene Compounds, William J. Robinson Iii

Browse all Theses and Dissertations

Separately, N-Heterocyclic carbene and Tetrathiafulvalene compounds show great promise for a variety of applications. N-heterocyclic carbenes exhibit desirable metal bonding characteristics which allow for modulation of the metal center via back-bonding interactions. Additionally, this creates a system of increased bonding strength creating stable complexes. Tetrathiafulvalene exhibits strong redox-active and optical characteristics. These are owed to an increased aromaticity as oxidation of the compound occurs and thus, is stable through a multitude of oxidative states. The marriage of these two functionalization creates a unique opportunity for a “cure-all” compound, able to address many problems the world faces today including, but not …

Characterization Of O-Linked Glycosylated Neuropeptides In The American Lobster (Homarus Americanus): The Use Of Peptide Labeling Following Beta Elimination, Edward Myron Bull Jan 2020

Characterization Of O-Linked Glycosylated Neuropeptides In The American Lobster (Homarus Americanus): The Use Of Peptide Labeling Following Beta Elimination, Edward Myron Bull

Honors Projects

Neuropeptides are a class of small peptides that govern various neurological functions, and the American lobster (Homarus americanus) provides a model system for their characterization. Neuropeptides are commonly post-translationally modified (PTM), and one common PTM is glycosylation. Past research in the Stemmler lab has found glycosylated neuropeptides in H. americanus; however, the extent and biological role of this modification has not been well characterized. This study was undertaken to determine the number of glycosylated peptides in the sinus glands of H. americanus and to develop an approach to tag the site of glycosylation using beta-elimination chemistry. LC-MS …

Cholinesterase Based System For Fentanyl Detection, Alyssa Shoemaker Jan 2020

Cholinesterase Based System For Fentanyl Detection, Alyssa Shoemaker

Legacy Theses & Dissertations (2009 - 2024)

Opioids and other illicit drugs have become a very prevalent problem across the world, especially in the United States. In fact, every day more than 130 people die from an opioid overdose. Despite many public warnings explaining the risks of opioid and drug use, the epidemic has continued to grow. As illicit drugs are becoming more rampant in society, law enforcement officers need a reliable, rapid, and non-invasive method to detects when someone may be on drugs, for example while driving, and to assess unknown powders or substances when presented at a crime scene containing drugs. Currently, at traffic stops …

Optimal Excitation Wavelength For Raman Spectroscopic Analysis Of Organic Gunshot Residue Evidence, Ashley Hull Jan 2020

Optimal Excitation Wavelength For Raman Spectroscopic Analysis Of Organic Gunshot Residue Evidence, Ashley Hull

Legacy Theses & Dissertations (2009 - 2024)

Gunshot residue (GSR) is a type of trace evidence that can often be recovered at violent crime scenes. There are two major types of GSR, organic gunshot residue (OGSR), and inorganic gunshot residue (IGSR). Traditional forensic analysis of GSR focuses on IGSR, although with the movement towards lead-free, or non-toxic ammunition, IGSR analysis has become less informative, as lead is a major component of IGSR analysis. Because of this, OGSR has been investigated to supplement the standard IGSR analysis methods. Previous work has shown Raman spectroscopy to be effective at identifying OGSR as well as a minimally destructive and specific …

Targeting Oncogenic K-Ras And Monoamine Oxidase B Utilizing Chalcones Bearing The 3,4,5-Trimethoxyphenyl Motif, Cody Fourman

Browse all Theses and Dissertations

Oncogenic K-Ras is a driver protein found in specific cancers and was once thought to be “undruggable.” However, recent advances have suggested that mislocalizing K-Ras from the plasma membrane could be a viable strategy to target these cancers. The objective of this project was to identify a lead compound and determine a structure-activity relationship from chalcones bearing the 3,4,5-trimethoxy motif towards oncogenic K-Ras and MAO-B. This was accomplished by keeping the trimethoxy group consistent on one ring while varying the electron-donating or -withdrawing groups on the opposite ring. Previous work showed chalcones of this specific archetype exhibiting indiscriminate MAO inhibition, …

Investigating Heterogeneous Nucleation Of Barite Using Hydrothermal Atomic Force Microscopy And Optical Microscopy, Ankita B. Gurav Jan 2020

Investigating Heterogeneous Nucleation Of Barite Using Hydrothermal Atomic Force Microscopy And Optical Microscopy, Ankita B. Gurav

Browse all Theses and Dissertations

In order to develop a better understanding heterogeneous nucleation of barite, barite precipitation was studied under varying experimental parameters. Hydrothermal atomic force microscopy (HAFM) and optical microscopy were used to investigate the effect of change in temperature, supersaturation and varying ratios of ions on heterogeneous nucleation of barite. In the experiments conducted at higher temperatures, the particles thus nucleated were found to display characteristic hexagonal and rhomboidal shapes. In comparing results of particle densities among different ion ratios, there is evidence suggesting that barium to sulfate ratio plays a role of promoter. Wherein, the ratios with higher [Ba2+] concentration were …
