Browse all Theses and Dissertations

The synthesis of poly(arylene ether)s, with a pendent aryl iodide, was achieved via nucleophilic aromatic substitution (NAS) polycondensation of DI-DFDPS monomer. The platform diiodo-difluorodiphenyl sulfone monomer, DFDPS-DI (3), was prepared in one-step, via EAS chemistry. Structural characterization was provided by NMR spectroscopy, GC/MS, and elemental analysis. As “pre” approach didn’t work here, so “post” approach was used to synthesize N-heterocycle functionalized copolymers. The DI-copolymers were subjected to copper-catalyzed C-N coupling reaction with carbazole, phenoxazine, and indole and the syntheses of chromophore based copolymer were successfully carried out. A series of PAEs containing carbazole, phenoxazine, and indole substituents were synthesized and …

Elephant And Anchors ‒ Photoelectron Photoion Coincidence Spectroscopy Of Small Oxygenated Molecules, Apeter Weidner

University of the Pacific Theses and Dissertations

The dissociative photoionization reactions of two small, oxygenated organics, namely 1,3-dioxolane and methyl vinyl ketone, were studied by photoelectron photoion coincidence (PEPICO) spectroscopy. Experiments involving 1,3-dioxolane were carried out in the photon energy range of 9.5‒13.5 eV. The statistical thermodynamics model shows that a total of six dissociation channels are involved in the formation of three fragment ions, namely C3H5O2+ (m/z 73), C2H5O+ (m/z 45) and C2H4O+ (m/z 44), with two channels contributing to the formation of each. By comparing the results of ab initio quantum chemical calculations to the experimentally derived appearance energies of the fragment ions, the most …

Honors Theses

G protein-coupled receptors (GPCRs) play a vital role in cell signaling, and imaging the internalization of these proteins will allow for better understand of protein trafficking within the cell, as well as identification of potential drug targets. However, there is a need for further development of fluorescent protein labels that are able to effectively monitor GPCR internalization in the near-infrared (NIR) range. We recently described a series of NIR fluorophores in which a phosphinate functionality is inserted at the bridging position of the xanthene scaffold, called the Nebraska Red (NR) dye series. These dyes are attractive reagents for the imaging …

Browse all Theses and Dissertations

A novel organic TADF chromophore was synthesized via a 3-step process to emit a blue fluorescent light upon excitation. The chromophore was covalently attached to a 61% isobutyryl amidated poly(ethyleneimine) (PEI-IBAm0.61) non-viral gene vector utilized in gene transfection experiments. The gene vectors are loaded with a green fluorescent protein (GFP) and are monitored via fluorescence microscopy. The addition of the blue chromophore therefore allows for the monitoring of the movements of the PEI-IBAm0.61 through the full delivery mechanism of the GFP inside the cells. This can give vital information about the overall toxicity of these non-viral gene vectors to biological …

Development Of A Computer Algorithm For Generation Of Primers For Nucleic Acid Sequence Based Amplification (Nasba), Rohit Karnati

Honors Undergraduate Theses

Nucleic acid sequence based amplification (NASBA) is a primer based isothermal method of RNA/DNA amplification. Currently, primer design for NASBA has been restricted to hand creating sequences of oligonucleotides that must follow a set of rules to be compatible for the amplification process. This process of hand-creating primers is prone to error and time intensive. The detection of mutants, post amplification, also offers a benefit in point of care scenarios and the design of hybridization probes for sequences in the region of amplification is also an erroneous and time intensive process. By creating a program to design primers and hybridization …

Cell-Trappable Chemiluminescent Probes For Monitoring Hydrogen Sulfide In Living Cells, Briley Bezner Dec 2019

Cell-Trappable Chemiluminescent Probes For Monitoring Hydrogen Sulfide In Living Cells, Briley Bezner

Chemistry Theses and Dissertations

Hydrogen sulfide (H2S) is an important biological signaling molecule that has been recognized alongside nitric oxide and carbon monoxide as being a small gasotransmitter that is enzymatically produced and impacts multiple physiological functions. To detect hydrogen sulfide, there has been a focus on developing fluorescent probes to target particular analytes; however, fluorescent probes lack sensitivity and depth penetration due to background autofluorescence and light scattering. Chemiluminescence does not require light excitation, which greatly reduces the amount of autofluorescence and photoactivation.

In order to detect hydrogen sulfide in living systems with high sensitivity, a series of sterically stabilized 1,2-dioxetane chemiluminescent reduction-reaction …

Design And Synthesis Of Tlr2 And Tlr6 Heterodimer Ligands, A Triply Functionalized Α-Galcer Derivative For Identifying Proteins Involved In Glycolipid Trafficking, And The Disaccharide Of Staphylococcus Aureus Cp8 Towards A Self-Adjuvanting Vaccine, Sara Mayeth Mata

Theses and Dissertations

Toll like receptors (TLRs) are found on B cells, macrophages, monocytes, and dendritic cells, and these cells belong to the innate immune system that recognizes antigens and induces multiple cell responses through the release of cytokines. TLR1, TLR2 and TLR6 function as heterodimers, either as TLR1/TLR2 or TLR2/TLR6 to recognize lipopeptides. TLR1/2 dimer activation releases inflammatory cytokines, while TLR2/TLR6 dimer activation releases immunomodulatory cytokines. Based on the size of the binding pocket between TLR2 and TLR6, it was hypothesized that lipopeptides, such as FSL1, could be simplified while keeping overall activity. FSL1 is a lipopeptide first isolated from Mycoplasma salivarum …
