The discovery of the atom and its associated components and properties paved the way for laying the foundation of chemistry related knowledge. However, the historical discoveries of each individual component are often forgotten or ignored due to the students‰Ûª innate belief of it ‰Û÷being difficult‰Ûª to remember such specific information. To assist students with grasping the properties of the atom and its components, a timeline-assisted chronology of atomic discoveries was created. Access to experimental X-ray diffraction image data is fundamental for validation and reproduction of macromolecular models and indispensable for development of structural biology processing methods. Here, we established a diffraction data publication and dissemination system, Structural Biology Data Grid (SBDG; data.sbgrid.org), to preserve primary experimental data sets that support scientific publications. Data sets are accessible to researchers through a community driven data grid, which facilitates global data access. Our analysis of a pilot collection of crystallographic data sets demonstrates that the information archived by SBDG is sufficient to reprocess data to statistics that meet or exceed the quality of the …

Ferrocene containing N-heterocyclic carbene (NHC) ligated gold(I) complexes of the type [Au(NHC)2]+ were prepared and found to be capable of regulating the formation of reactive oxygen species (ROS) via multiple mechanisms. Single crystal X-ray analysis of bis(1-(ferrocenylmethyl)-3-mesitylimidazol-2-ylidene)-gold(I) chloride (5) and bis(1,3-di(ferrocenylmethyl)imidazol-2-ylidene)-gold(I) chloride (6) revealed a quasi-linear geometry around the gold(I) centers (i.e., the C–Au–C bond angle were measured to be ∼177° and all the Au–Ccarbene bonds distances were in the range of 2.00 (7)–2.03 (1) Å). A Novel Dual Allosteric Activation Mechanism Of Escherichia Coli Adp-Glucose Pyrophosphorylase: The Role Of Pyruvate, Matías D. Asención Diez, Mabel C. Aleanzi, Alberto A. Iglesias, Miguel Ballicora
Miguel A Ballicora
Fructose-1,6-bisphosphate activates ADP-glucose pyrophosphorylase and the synthesis of glycogen in Escherichia coli. Here, we show that although pyruvate is a weak activator by itself, it synergically enhances the fructose-1,6-bisphosphate activation. They increase the enzyme affinity for each other, and the combination increases Vmax, substrate apparent affinity, and decreases AMP inhibition. Our results indicate that there are two distinct interacting allosteric sites for activation. Hence, pyruvate modulates E. coli glycogen metabolism by orchestrating a functional network of allosteric regulators. We postulate that this novel dual activator mechanism increases the evolvability of ADP-glucose pyrophosphorylase and its related metabolic control.

Teaching Climate Change Concepts And The Nature Of Science: A Library Activity To Identify Sources Of Climate Change Misconceptions, Charity Flener Lovitt, Kristen S. Shuyler

A library activity was developed in which students found information about climate science misconceptions from popular and scientific literatures. As part of the activity, students developed a rubric to evaluate the credibility and type of literature sources they found. The activity prepared students to produce an annotated bibliography of articles, which they then used to create a training document about a climate science misconception for staff at a local science center. Evaluation of annotated bibliographies showed that students were able to distinguish between popular and scholarly literature but struggled to identify primary and secondary sources within the scholarly literature. Sialic acids (SAs) are a diverse family of naturally occurring 2-keto-3-deoxy-nononic acids that are involved in a wide range of biological processes, including early fetal development, cellular recognition, and utilization by microbes. While it is clear that cell surface SAs are highly involved in the immune system, the sialylation status of individual immune cells and functions are still unknown. In this study, I combined the newly developed LC-MS/MS methods with flow cytometry and confocal microscopy to systematically study the sialylation and desialylation dynamics of macrophages at different conditions.

Assignments in which students edit Wikipedia may help students learn about the complexities of information creation and production, while engaging them in researching and writing about topics related to class content. This chapter presents two case studies that illustrate how Wikipedia-based activities can be designed to achieve both chemistry and information literacy learning outcomes. In both examples, faculty partnered with a librarian to implement the Wikipedia editing assignments. Through these experiences, those involved learned about Wikipedia and its community, and identified promising practices for project requirements based on formal and informal assessment and observations. Forensic laboratories face issues with large backlogs on evidence needing to be analyzed is due to two reasons: budgeting issues and the emergence of designer drugs. Currently, forensic laboratory directors cite that there are a shortage of scientists to deal with the yearly increase in caseloads. Additionally, designer drugs like synthetic cathinones, commonly known as “bath salts,” have increased the backlog due to the changing chemical compositions and misidentification as other compounds. Therefore, the use of a portable mass spectrometer for on-site forensic chemical analysis of synthetic cathinones would reduce the backlog of suspected controlled substances in forensic chemistry laboratories. …

The synthesis and fluorescent analysis of chiral light emitting ionic liquids made from lanthanide-doped chiral amino acid ionic liquids is described in this thesis. The ionic liquids were composed of tetrabutylammonium (TBA) cation with enantiomers of the amino acids valine, proline, and aspartate as anionic counterions. These ionic liquids of varying amino acid composition were doped with europium trifluoromethanesulfonate (triflate) and analyzed by circularly polarized luminescence (CPL). The sign of the CPL signal corresponded to the handedness of the amino acids in the ionic liquids. Europium triflate concentration dependent CPL in both [TBA][Val] and [TBA][L-Pro] ionic liquids was measured from …

A mixture of vanadium(V) oxide and copper sulfate was used to catalyze oxidative transformations in a variety of organic compounds. Multiple functionalities were tested, including thiols, amines and alcohols. Various transformations were observed resulting in various oxidized products, including coupling reactions. Reactions were performed in a solvent-free environment due to the insoluble nature of vanadium(V) oxide in common organic solvents. Results were analyzed using HNMR and GCMS.

Mycobacterium ulcerans is the causative agent of Buruli ulcer, a tropical skin disease that affects thousands of individuals annually. Recent studies have revealed that lipolytic enzymes are involved in the pathogenicity processes of mycobacterium and could be potential targets for novel antibiotics. LipN is one proposed serine hydrolase in Mycobacterium ulcerans that contains the conserved α/β hydrolase protein fold and utilizes the conserved catalytic traid of serine, histidine, and aspartate/glutamate. This dissertation focuses on the synthesis of dauer pheromone analogs for biological evaluation, the development of visible light promoted O-glycosylation methods and the development of an Ir(ppy)3 catalyzed C-H hydroxylation method. Chapter 1 describes the synthesis of dauer pheromone analogs. C.elegans is a small nematode that enters a dauer stage when encountering unfavorable environmental conditions. The dauer stage is initiated by the nematodes chemosensation of the dauer pheromone caused by the down regulation biochemical pathways TGF-â and IGF-1. These biochemical pathways are also seen in Homo Sapiens and play a role in a number of biological processes. Understanding how these …

On October 7, 2015, the Nobel Prize in Chemistry for 2015 was awarded to three deserving scientists for their pioneering research on DNA repair. Tomas Lindahl was recognized for studies that uncovered the inherent instability of DNA as well as the mechanism of the base excision repair pathway, Paul Modrich for characterization of the mismatch repair pathway, and Aziz Sancar for mechanistic elucidation of the nucleotide excision repair (NER) pathway. Characterizing the interactions of solvent molecules with ions is fundamental in understanding the thermodynamics of solution chemistry. These interactions are difficult to observe directly in solution because the number of solvent molecules far exceed that of ions. This lend the gas phase to be the ideal medium in the study ion-solvent interactions on a molecular level. Ionized polycyclic aromatic hydrocarbon (PAH) molecules can readily form hydrogen bonds with neutral solvent molecules in aqueous and interstellar medium. Various 1,3,4-oxadiazol-2(3H)-ones were synthesized by reaction of the bromocarbonylhydrazine salt(35) obtained from treatment of 4-bromo-3-phenylsydnone(34) with HCl with the corresponding acid chlorides in DME. The structures were confirmed by their physical, analytical and spectral data (IR, GC/MS, 1H and 13C NMR and CHN analysis). In general these compounds were synthesized in good to excellent yields and it is planned to continue exploration of their chemical behavior as a guide to their potential antimicrobial activity. Poly(arylene ether sulfone)s with pendent aryl iodide group was successfully synthesized via Nucleophilic aromatic polycondensation of 1-(3,5-difluorophenyl sulfonyl)-4-iodobenzene using 4,4’-difluorodiphenylsulfone (DFDPS) and Bisphenol A in the ratio 25:75:100. Iodo monomer was prepared by reacting 3,5-difluorodiphenyl sulfone with N-iodo succinimide (NIS). Using GC/MS and NMR spectroscopy, the quantitative analysis was done to this monomer. Modification was done to iodo copolymer using sodium azide and copper sulfate as catalyst to form azide copolymer. The proliferation of silver nanoparticles (AgNPs) in consumer products and industrial applications has generated many environmental concerns since the chemistry of silver changes at the nanoscale and the very properties that make them desirable – their unique reactivity – may pose environmental risks. This dissertation addresses these concerns from three different angles. First, freshwater crayfish is demonstrated as a potential benthic indicator of Ag pollution since Ag accumulation depends on the form of Ag. The vast majority of emissions of fluorine-containing molecules are anthropogenic in nature, e.g. chlorofluorocarbons (CFCs), hydrochlorofluorocarbons (HCFCs), and hydrofluorocarbons (HFCs). Many of these fluorine-containing species deplete stratospheric ozone and are regulated by the Montreal Protocol. Once in the atmosphere they slowly degrade, ultimately leading to the formation of hydrogen fluoride (HF), the dominant reservoir of stratospheric fluorine due to its extreme stability. Monitoring the growth of stratospheric HF is therefore an important marker for the success of the Montreal Protocol. Antioxidants are a class of molecules that provide a protective function against reactive oxygen species (ROS) in biological systems by out competing physiologically important molecules for ROS oxidation. In natural waters, the reactivity of antioxidants gives an estimate of oxidative stress, and may determine the reactivity and distribution of reactive oxidants. We present an analytical method to measure antioxidant activity in natural waters through the competition between ascorbic acid, an antioxidant, and MCLA, a chemiluminescent probe for superoxide. A numerical kinetic model of the analytical method has been developed to optimize analytical performance. Measurements of antioxidant concentrations in pure and …
