Hyper-immunoglobulin E syndrome (HIES) is a primary immune deficiency characterized by abnormal and devastating susceptibility to a narrow spectrum of infections, most commonly Staphylococcus aureus and Candida albicans. Recent investigations have identified mutations in STAT3 in the majority of HIES patients studied. Despite the identification of the genetic cause of HIES, the mechanisms underlying the pathological features of this disease remain to be elucidated. Here, we demonstrate a failure of CD4+ T cells harboring heterozygous STAT3 mutations to generate interleukin 17-secreting (i.e., T helper [Th]17) cells in vivo and in vitro due to a failure to express sufficient levels of …

Background: Mediastinitis resulting from surgical site infection may occur in 1% of patients undergoing median sternotomy. Methods: Case report and review of the pertinent English-language literature. Results: We report a case of mediastinitis caused by Acinetobacter baumannii, in a patient with multiple comorbidities who underwent cardiopulmonary bypass. Successful treatment consisted of surgical debridement, reconstruction, and ampicillin-sulbactam. Conclusions: Acinetobacter baumannii should be recognized as a potential causative agent of severe postoperative mediastinitis.

Diagenesis is now recognized as a potentially major source of error in paleoclimatic reconstructions from fossil and modern coral geochemical records. Key to avoiding spurious results caused by diagenesis is thorough screening of coral material prior to geochemical analysis. In this data brief we present color images from thin sections of fossil and modern Porites corals and demonstrate the effectiveness of thin sections in detecting low levels of diagenesis. The images presented here cover a range of coral preservation levels from pristine aragonite to 100% calcite. New results of CO global total column measurements using the Atmospheric Infrared Sounder (AIRS) aboard the Aqua satellite in comparison with Measurements of Pollution in the Troposphere (MOPITT) sensor aboard the Terra satellite are presented. Both data sets are validated using ground-based total column measurements in Russia and Australia. A quality parameter based on the Profile Percent A Priori values from the standard MOPITT product is introduced. AIRS data (version 4) for biomass burning events are in agreement or lower than both MOPITT and ground measurements, but CO bursts can be seen by AIRS in most cases. For the cases …

The impact of an exotic species in natural systems may be dependent not only on invader attributes but also on characteristics of the invaded community. We examined impacts of the invader bitou bush, Chrysanthemoides monilifera ssp. rotundata , in fore and hind dune communities of coastal New South Wales, Australia. We compared invader impacts on vegetation structure, richness of both native and exotic growth forms and community variability in fore and hind dunes. We found that impacts of bitou invasion were context specific: in fore dune shrublands, functionally distinct graminoid, herb and climber rather than shrub growth forms had significantly …

Ultrafine fibers with diameters ranging from 50 to 300 nm were prepared from DNA/single-walled carbon nanotubes (SWNTs)/poly(ethylene oxide) blended dispersion. Well-defined electrospun fibers were obtained by good control of key dispersion properties related to electrospinning, such as ionic conductivity, surface tension, and viscosity. Raman spectroscopy confirmed the presence of SWNT in the resulting fibers, indicating good interaction between DNA and SWNT. The resulting fibers also exhibited electroactive behavior and could be used as an immobilization matrix for a glucose oxidase enzyme biosensor. The electro-deposition of titanium from two different classes of ionic liquids has been studied. Gold ions in solution have been used as an additive in order to provide a stable nucleation site for titanium to deposit on via a co-deposition process. Despite evidence from previous literature, it was found that it was not possible to reduce titanium from the +4 state to titanium (0) due to problems of redox cycling and solubility of reduced titanium species in the various ILs trialled. Trilayer polypyrrole benders are capable of generating voltages and currents when applied with an external force or displacement, demonstrating potential as mechanical sensors. Previous work has identified the effects of dopant and electrolyte on the sensor output, and a 'deformation induced ion flux' model was proposed. The current work aims to identify the change in sensor response with input amplitude and bender geometry as a function of frequency. The current and charge output from the trilayer benders were found to increase proportionally with input displacement and bender strain for multiple input frequencies, indicating linearity. Graphene sheets offer extraordinary electronic, thermal and mechanical properties and are expected to find a variety of applications. A prerequisite for exploiting most proposed applications for graphene is the availability of processable graphene sheets in large quantities. The direct dispersion of hydrophobic graphite or graphene sheets in water without the assistance of dispersing agents has generally been considered to be an insurmountable challenge. Here we report that chemically converted graphene sheets obtained from graphite can readily form stable aqueous colloids through electrostatic stabilization. The controlled deposition of polyaniline carbon nanotube composites by inkjet printing is reported. It is demonstrated that the sheet resistance and transmittance can be expressed in amount of composite and MWNT material deposited. The most efficient way for improving the sheet resistance while keeping the cost in optical transparency to a minimum is by increasing the total amount of material deposited, rather than increasing MWNT loading fraction.

Hydrogen chloride (HCl) and hydrogen fluoride (HF) are respectively the main chlorine and fluorine reservoirs in the Earth's stratosphere. Their buildup resulted from the intensive use of man-made halogenated source gases, in particular CFC-11 (CCl3F) and CFC-12 (CCl2F2), during the second half of the 20th century. It is important to continue monitoring the evolution of these source gases and reservoirs, in support of the Montreal Protocol and also indirectly of the Kyoto Protocol. In late December 2003, a wildfire in the Jervis Bay region of New South Wales burned through an area that previously supported a large population of the endangered eastern bristlebird (Dasyornis brachypterus). The eastern bristlebird has been described as fire-sensitive, and fire is implicated in the decline of the species. The frequency of occurrence of bristlebirds was investigated in the second week after the fire in a range of sites varying in fire intensity. Bristlebirds were found in burned habitats but were less common in the sites that were more intensely burnt. Bristlebirds had been surveyed along transects in this …

Cosmomycin D (CosD) is an anthracycline that has two trisaccharide chains linked to its ring system. Gel electrophoresis showed that CosD formed stable complexes with plasmid DNA under conditions where daunorubicin (Dn) and doxorubicin (Dx) dissociated to some extent during the experiments. The footprint and stability of CosD complexed with 10- and 16 mer DNA was investigated using several applications of electrospray ionisation mass spectrometry (ESI-MS). ESI-MS binding profiles showed that fewer CosD molecules bound to the sequences than Dn or Dx. In agreement with this, ESI-MS analysis of nuclease digestion products of the complexes showed that CosD protected the …

Reconstructing the phylogeny and biogeography of the Caribbean land snail Cerion requires a robust stratigraphic and chronological framework. To this end, we have determined the stratigraphic succession on San Salvador, a Bahamian island with a rich fossil and modern Cerion fauna. A primary purpose of this paper is to independently verify this succession through whole-rock and Cerion aminostratigraphies and AMS 14C-based age models. Over 150 individual Cerion shells were age-ranked from 140 ka to modern using stratigraphic position and reverse-phase HPLC (RPC) amino acid racemization, which was sufficiently sensitive to resolve stratigraphic subunits within the Holocene and late Pleistocene. …

Species distribution models (SDMs) often use elevation as a surrogate for temperature or utilise elevation sensitive interpolations from weather stations. These methods may be unsuitable at the landscape scale, especially where there are sparse weather stations, dramatic variations in exposure or low elevational ranges. The goal of this study was to determine whether radiation, moisture or a novel estimate of exposure could improve temperature estimates and SDMs for vegetation on the Illawarra Escarpment, near Sydney, Australia. Tumour expression of the urokinase plasminogen activator correlates with invasive capacity. Consequently, inhibition of this serine protease by physiological inhibitors should decrease invasion and metastasis. However, of the two main urokinase inhibitors, high tumour levels of the type-1 inhibitor actually promote tumour progression, whereas high levels of the type-2 inhibitor decrease tumour growth and metastasis. We propose that the basis of this apparently paradoxical action of two similar serine protease inhibitors lies in key structural differences controlling interactions with components of the extracellular matrix and endocytosis/signalling co-receptors.

Siliceous inclusions play an important role in deterring terrestrial herbivores, but their role in marine systems remains controversial. Sponges are ubiquitous members of marine benthic communities and represent a potentially valuable food source for many predatory species, yet they have few natural predators. An in situ method was used to assess the deterrent effects of siliceous spicules or whole sponge skeleton, which were hypothesised to act as a physical defense in temperate reef sponges. Optimal debulking followed by chemotherapy is the standard treatment of managing late-stage ovarian cancer, but chemoresistance is still a major problem. In this study, we compared expression profiles of primary tumor tissue from five long-term (>8 years) and five short-term (years) ovarian cancer survivors and identified clusterin as one of the genes that were significantly up-regulated in short-term survivors. We then evaluated the prognostic significance of clusterin and its possible correlation with chemoresistance in ovarian cancer by immunohistostaining of clusterin in 62 tumor samples from patients with stage III, high-grade serous ovarian cancer. The pathology of more than 40 human degenerative diseases is associated with fibrillar proteinaceous deposits called amyloid. Collectively referred to as protein deposition diseases, many of these affect the brain and the central nervous system. In many cases the amyloid deposits are extracellular and are found associated with newly identified abundant extracellular chaperones (ECs). Evidence is discussed which suggests an important regulatory role for ECs in amyloid formation and disposal in vivo. This is emerging as an exciting field. Within the European project UFTIR (Time series of Upper Free Troposphere observations from an European ground-based FTIR network), six ground-based stations in Western Europe, from 79° N to 28° N, all equipped with Fourier Transform infrared (FTIR) instruments and part of the Network for the Detection of Atmospheric Composition Change (NDACC), have joined their efforts to evaluate the trends of several direct and indirect greenhouse gases over the period 1995–2004. This paper describes the statistical analysis of annual trends in long term datasets of greenhouse gas measurements taken over ten or more years. The analysis technique employs a bootstrap resampling method to determine both the long-term and intra-annual variability of the datasets, together with the uncertainties on the trend values.

Research over the past two decades has provided significant epidemiological and other evidence for the health benefits of the consumption of soy-based foods. A large number of dietary intervention studies have examined the effects of soy isoflavones on risk factors for cardiovascular disease and hormone-dependent cancers. However, these report large variability in outcome measures, very limited reproducibility between studies and in some cases, controversy between results of clinical trials using dietary soy or soy protein and isoflavone supplementation. -Textile based sensors which can be used to measure the chemical composition of bodily fluids represents a major advancement in the area of wearable technology. BIOTEX is an EU funded project aiming to develop such sensors with a particular interest in monitoring perspiration. A textile based fluid handling system has been developed for sample collection and transport. Sodium, conductivity and pH sensors have also been developed. This paper details the integration and testing of these sensors.
