C. albicans is a polymorphic fungal pathogen which has the ability to shift from yeast to hyphae. C. albicans cell wall is composed of glucan, chitin, mannoprotein and mannan. It is not possible, using standard extraction methods, to isolate mannan from C. albicans hyphae. To isolate hyphal mannan, we developed a simplified alkali extraction method. Using this method it was determined that hyphal mannan has a much lower molecular weight, a smaller polymer distribution and altered conformation structure when compared to yeast mannan. According to the CDC, the most common type of heart disease is coronary artery disease, which frequently leads to myocardial infarction (MI). Therapeutic approaches to lessen the resulting cardiovascular injury associated with MI are limited. Recently, the management paradigm for cardiac injury has entered the molecular era and microRNAs (miRNAs) have been shown to act as negative regulators of gene expression by inhibiting mRNA translation and/or stimulating mRNA degradation. A single miRNA can modulate physiological or disease phenotypes by regulating whole functional systems. With the development of a strategy to correlate the dark-field light scattering spectra of individual nanostructures with scanning electron microscopy (SEM) and atomic force microscopy (AFM) images of the same nanostructures, we were able to investigate several interesting optical properties of Au nanoplates (NPs) and asymmetrically-coupled Au nanospheres (NSs) attached to Au NPs with a high level of control. The light scattering spectra of the NP/NS coupled structures depend strongly on the location of NS attachment on the NP. The bright eclipsing binary system epsilon Aurigae offers a unique opportunity to uncover physical mechanisms contributing to disk formation and evolution and to explore the relationship between photospheric anisotropies, stellar pulsation and mass loss. This research contributes to our understanding of stellar evolution in the context of binary stars. The current dissertation focuses on the effects of sulfur‒oxidation on nitrile hydration activity of a series of ruthenium(II) complexes inspired by nitrile hydratase (NHase). NHase catalytically hydrates nitriles to amides at a low spin non‒heme Fe(III) or non‒corrin Co(III) metal center. The active‒site employs a N2S3X type donor set using two carboxamido nitrogens and three cofacial cysteinic sulfurs that are present in distinct oxidation states as sulfinate (RSO2¯), sulfenate (RSO¯), and thiolate (RS¯). The distance d(u, v) between two vertices u and v in a graph G equals the length of a shortest path from u to v. A set S of vertices is called a distance-2 dominating set if every vertex in V \S is within distance-2 of at least one vertex in S. The distance-2 domatic number is the maximum number of sets in a partition of the vertices of G into distance-2 dominating sets. We give bounds on the distance-2 domatic number of a graph and determine the distance-2 domatic number of selected classes of graphs.

Clark’s Cave contains a large collection of late Pleistocene mammal material. In particular, it contains a sizable amount of Microtus spp. which can be valuable paleoclimate indicators. Identification techniques traditionally used to classify these species have been shown to be unreliable. Recent studies have shown that using geometric morphometric techniques on lower first molars can be more successful. By placing landmarks and running a discriminate analysis on new and previously collected material from the cave, significant differences in Microtus species proportions were found. The American alligator, Alligator mississippiensis, is a large extant archosaur and member of the Order Crocodilia. Crocodilian ontogeny has been studied in great detail, the skull being of particular interest. One aspect of the skull left unstudied is how individual cranial elements change through ontogeny independent of one another. This study observed morphological change in a growth series of 34 specimens of A. mississippiensis from ETSU Vertebrate Paleontology Lab collections. The premaxilla, maxilla, nasal, jugal, frontal, and parietal were analyzed using landmark morphometrics. The frontal, jugal, and parietal showed more allometric growth with the orbits reducing in size posteriorly. …

The Pile Mound survey includes magnetometry paired with targeted ground-penetrating radar (GPR) and electromagnetic induction (EMI) surveys of the mound and testing of associated features over the ca. 6.5 ha site. The GPR survey discovered six rock features (five large rock features within the mound and one marking the outside of the mound). Knowledge of mounds in the Upper Cumberland Plateau (UCP) is lacking—the closest other studied sites are at the Corbin Site, Croley-Evans, Bell Site, and Beasley Mounds, approximately 75 – 100 km away. One of the most prevalent inherited diseases is cystic fibrosis. This disease is caused by a mutation in a membrane protein, the cystic fibrosis transmembrane conductance regulator (CFTR). CFTR is known to function as a chloride channel that regulates the viscosity of mucus that lines the ducts of a number of organs. Generally, most of the prevalent mutations of CFTR are located in one of two nucleotide binding domains, namely, the nucleotide binding domain 1 (NBD1). The Gray Fossil Site (GFS), a Mio-Pliocene (4.5–7 Ma) locality in the southern Appalachians, boasts the most diverse pre-Pleistocene salamander fauna in North America: Desmognathus sp., Plethodon sp., Notophthalmus sp., a Spelerpinae-type plethodontid, and Ambystoma sp. Because greater taxonomic resolution can result in more precise paleobiological interpretations, additional specimens were studied here. ETMNH 8045, a nearly complete articulated ambystomatid, appears most like Ambystoma maculatum in dentition and vertebral proportions. ETMNH 18219, an isolated vomer, is consistent with modern Pseudotriton and Gyrinophilus in possessing a postdentigerous process and a similar dentigerous row morphology. If these taxa, or species …

Vitamin E exhibits anti-tumor activity by regulating pathways in cancer cells, potentially the lipoxygenase (LOX) pathway. We studied the effects of alpha tocopherol (AT), gamma tocopherol (GT), gamma tocotrienol (GT3), and an alpha-gamma tocopherol mixture (ATGT) on the production of the LOX metabolites 13-hydroxyoctadecaenoic acid (HODE), 15-hydroxyeicosatetraenoic acid (HETE), 12-HETE, and 5-HETE in colorectal cancer. These metabolites were examined in the HCT-116 cell line after 24 h treatment with select vitamin E isoforms and quantified by LC/MS/MS. Six new crystal structures were determined using a Rigaku Mercurcy 375/MCCD(XtaLab mini) diffractometer. The structure of a molybdenum cofactor was solved resulting in an R1 (R1 = Σ ||Fo| - |Fc|| / Σ |Fo|) of 3.61% despite the presence of a disordered DMSO molecule. New Tetrakis(N-phenylacetamidato) Dirhodium(II) complexes were synthesized and characterized. Two 2,2-cis-[Rh2(NPhCOCH3)4]•(C3H4N2)x where x= 1 or 2 were successfully crystallized and solved with R1 values below 5%. This thesis describes a study of the ligand chemistry of Pd Monolayer-Protected Clusters (MPCs) synthesized in the presence of octylamine. MPCs consists of a metal core surrounded by various organic stabilizers. Pd MPCs coated with alkanethiols, alkylamine, dendrimers, surfactants, and polymers have recently been explored for catalysis applications.Pd MPCs coated with alkylamines are more catalytically active than alkanethiols for hydrogentation/isomerization of allyl alcohol, but at shorter chain lengths are unstable in H2. In this document I describe a novel implementation of the generalized bisection method for finding roots of highly non-linear functions of several variables. Several techniques were optimized to reduce computation time. The implementation of the bisection method allows for the calculation of heterogeneous systems with SCED-LCAO, since derivative-based methods often fail for these systems. Systems composed of Gallium and Nitrogen are currently receiving much interest due to their behavior as semi-conductors and their ability to form nano-wires. Outliers are defined based on location only and also with respect to associated variables. Throughout the thesis we discuss five case studies, three of them come from experiments with spiders and bees, and the other two are data from earthquakes in a certain region. For aesthetic, economic, and health-related reasons, allowable concentrations of iron (Fe) and manganese (Mn) found present in drinking water are 0.3 mg/L and 0.05 mg/L, respectively. Water samples taken from private drinking wells in the rural communities within Buncombe County, North Carolina contain amounts of these metals in concentrations higher than the suggested limits. This study focused on bedrock geology, elevation, saprolite thickness, and well depth to determine factors affecting Fe and Mn. Using ArcGIS 10.2, spatial trends in Fe and Mn concentrations ranges were visualized, and estimates of the metal concentrations were interpolated to unmonitored areas. Results from this …

β-glucan is a major component of the fungal cell wall consisting of (1→3)-β linked glucose polymers with (1→6)-β linked side chains. The published classical isolation procedure of β-glucan from Saccharomyces cerevisiae is expensive and time-consuming. Thus, the aim of this research was to develop an effective procedure for the extraction of glucans. We have developed a new method for glucan extraction that will be cost effective and will maintain the native structure of the glucan. In this thesis we modify an existing model for the spread of resistant bacteria in a hospital. The existing model does not account for some of the trends seen in the data found in literature. Anacardic acids are salicylic acid derivatives known to possess various medicinal benefits. Gene silencing techniques, such as RNA interference, could be useful in determining specific enzymes involved in the synthesis of AnAcs. This dissertation introduces a technique for testing proper failure mitigation in safety critical systems. Unlike other approaches which integrate behavioral and failure models, and then generate tests from the integrated model, we build safety mitigation tests from an existing behavioral test suite, using an explicit mitigation model for which we generate mitigation paths which are then woven at selected failure points into the original test suite to create failure-mitigation tests (safety mitigation test).

There are three types which differ in their primary energetic materials; where single base powders contain nitrocellulose as their primary energetic material, double and triple base powders contain nitroglycerin in addition to nitrocellulose, and triple base powders also contain nitroguanidine.
