The lack of a rapid and quantitative autophagy assay has substantially hindered the development and implementation of autophagy-targeting therapies for a variety of human diseases. To address this critical issue, we developed a novel autophagy assay using the newly developed Cyto-ID fluorescence dye. We first verified that the Cyto-ID dye specifically labels autophagic compartments with minimal staining of lysosomes and endosomes. We then developed a new Cyto-ID fluorescence spectrophotometric assay that makes it possible to estimate autophagy flux based on measurements of the Cyto-ID-stained autophagic compartments. Anthropogenic atmospheric CO2 reacts with water to form carbonic acid (H2CO3) which increases water acidity. While marine acidification has received recent consideration, less attention has been paid to the effects of atmospheric carbon dioxide on freshwater systems—systems that often have low buffering potential. Since many aquatic systems are already impacted by pollutants such as heavy metals, we wondered about the added effect of rising atmospheric CO2 on freshwater organisms. We studied aquatic pulmonate snails (Physella columbiana) from both a heavy-metal polluted watershed and snails from a reference watershed that has not experienced mining pollution. Despite the importance of carbonate aquifers as reservoirs of oil, of water, and of carbon dioxide, questions remain about the diagenetic processes that enhance porosity and permeability. Some of the questions are: 1) what are the water-rock interactions that control the geochemistry of spring waters; 2) are the geochemical signatures of saline-fresh water mixing zones preserved in the carbonate rock record; and 3) how much carbon dioxide outgases along spring runs? Field-induced nanopore wetting by aqueous solutions, including electrolytes, provides opportunities for a variety of applications. Con!icting porosity requirements have so far precluded direct implementations of a two-way control: the pores have to be su"ciently wide to allow water in#ltration at experimentally relevant voltages but should not exceed the kinetic threshold for spontaneous expulsion in the absence of the #eld. Applicable widths are restricted below a few nanometers. Only a narrow window of #elds and pore geometries can simultaneously satisfy both of the above requirements. Hydride vapor phase epitaxy (HVPE) is used for the growth of low-defect GaN. We have grown undoped films on sapphire and investigated them using steady-state and time-resolved photoluminescence (PL). One of the dominant PL bands in high-quality GaN grown by HVPE is the green luminescence (GL) band with a maximum at 2.4 eV. This PL band can be easily recognized in time-resolved PL measurements due to its exponential decay even at low temperatures (<50 K), with a characteristic lifetime of 1–2 μs. As the temperature increases from 70 K to 280 K, the PL lifetime for the GL band increases by an order of magnitude. This …

Knowledge bases have been used to improve performance in applications ranging from web search and event detection to entity recognition and disambiguation. More recently, knowledge bases have been used to analyze social data. Wide adoption of smartphones and availability of low-cost sensors has resulted in seamless and continuous monitoring of physiology, environment, and public health notifications. However, personalized digital health and patient empowerment can become a reality only if the complex multisensory and multimodal data is processed within the patient context. Public institutions are increasingly reliant on data from social media sites to measure public attitude and provide timely public engagement. Such reliance includes the exploration of public views on important social issues such as gender-based violence (GBV). In this study, we examine big (social) data consisting of nearly fourteen million tweets collected from Twitter over a period of ten months to analyze public opinion regarding GBV, highlighting the nature of tweeting practices by geographical location and gender. Na-K-2Cl cotransporter 2A (NKCC2A), also known as the bumetanide-sensitive cotransporter 1 (BSC1), transports Na+, K+ and Cl- with a stoichiometry of 1:1:2. NKCC2A is considered a kidney specific cotransporter. It is abundantly expressed in apical membrane of the tubular cells in the thick ascending limb of the loop of Henle (TALH) and in the macula densa. However, NKCC2 has also been found at low levels in different cells, including insulin-secreting ones. Terahertz radiation is invaluable in spectroscopy and imaging due to its nondestructive nature. It has become a key focus for those wishing to develop sensors capable of detecting weapons and narcotics unobtrusively and at a distance. An ultrafast pulsed (femtoseconds) laser incident on a superconducting ring causes the emission of terahertz (THz) radiation. Commercial production of transparent conducting oxide (TCO) polycrystalline films requires high electrical conductivity with minimal degradation in optical transparency. Aqueous solution deposited TCO films would reduce production costs of TCO films but suffer from low electrical mobility, which severely degrades both electrical conductivity and optical transparency in the visible spectrum. Here, we demonstrated that grain boundary modification by ultra-violet laser crystallization (UVLC) of solution deposited aluminium-doped zinc oxide (AZO) nanocrystals results in high Hall mobility, with a corresponding dramatic improvement in AZO electrical conductance. A parameter used in the Black-Scholes equation, volatility, is a measure for variation of the price of a financial instrument over time. Determining volatility is a fundamental issue in the valuation of financial instruments. This gives rise to an inverse problem known as the calibration problem for option pricing. This problem is shown to be ill-posed. We propose a regularization method and reformulate our calibration problem as a problem of finding the local volatility in a reproducing kernel Hilbert space. Many types of invariants are used in the study of knots. Some are based on polynomials, some are purely algebraic, and some have their origins in geometry. One of the best known geometric knot invariants is the genus of a knot. A closely related but lesser-known invariant, crosscap number, was first introduced by Bradd Evans Clark in 1978. In mathematical epidemiology, disease transmission is commonly assumed to behave in accordance with the law of mass action; however, other disease incidence terms also exist in the literature. A homogeneous Susceptible-Infectious-Removed (SIR) model with a generalized incidence term is presented along with analytic and numerical results concerning effects of the generalization on the global disease dynamics. The spatial heterogeneity of the metapopulation with nonrandom directed movement between populations is incorporated into a heterogeneous SIR model with nonlinear incidence. The theory of stretching and compressing of short light pulses by the chirped volume Bragg gratings (CBG) is reviewed based on spectral decomposition of short pulses and on the wavelength-dependent coupled wave equations. The analytic theory of diffraction efficiency of a CBG with constant chirp and approximate theory of time delay dispersion are presented. Based on those, we performed comparison of the approximate analytic results with the exact numeric coupled-wave modeling. Coupled laser systems have application in everyday modern life, including for example, telecommunications and cryptography. Therefore it is important to gain insight into the dynamic processes at work and theoretical modeling can elucidate such processes. The work presented in this thesis is primarily concerned with aspects such as excitability and phase locking in mutually coupled oscillators. We compute the renormalized expectation value of the square of a massless, conformally coupled, quantum scalar field on the brane of a higher-dimensional black hole. Working in the AADD braneworld scenario, the extra dimensions are flat and we assume that the compactification radius is large compared with the size of the black hole. The four-dimensional on-brane metric corresponds to a slice through a higher-dimensional Schwarzschild-Tangherlini black hole geometry and depends on the number of bulk space-time dimensions. We examine a two-media 2-dimensional fluid system consisting of a lower medium bounded underneath by a flatbed and an upper medium with a free surface with wind generated surface waves but considered bounded above by a lid by an assumption that surface waves have negligible amplitude. An internal wave driven by gravity which propagates in the positive x-direction acts as a free common interface between the media. We call a hamiltonian N-space primary if its moment map is onto a single coadjoint orbit. The question has long been open whether such spaces always split as (homogeneous) x (trivial), as an analogy with representation theory might suggest. For instance, Souriau's barycentric decomposition theorem asserts just this when N is a Heisenberg group. For general N, we give explicit examples which do not split, and show instead that primary spaces are always flat bundles over the coadjoint orbit. In current debates about Buddhist approaches to the non-human natural world, studies describe Buddhism variously as anthropocentric, biocentric or ecocentric. These perspectives derive for the most part from examinations of philosophical and normative aspects of the tradition without much attention to moments when embodied practice diverges from religious ideals. Responding to the need for narrative thick descriptions of lived Buddhist attitudes toward nature, I ethnographically explore a Vietnamese monastery in the United States.
